Inhibition of Tanshinone IIA, Salvianolic Acid A and Salvianolic Acid B on Areca Nut Extract-Induced Oral Submucous Fibrosis in Vitro

Abstract: Salvia miltiorrhiza Bunge has been reported to possess excellent antifibrotic activity. In this study, we have investigated the effect and mechanism of tanshinone IIA1 (Tan-IIA1), salvianolic acid A (Sal-A) and salvianolic acid B (Sal-B), the important active compounds of Salvia miltiorrhiza Bunge, on areca nut extract (ANE)-induced oral submucous fibrosis (OSF) in vitro. Through human procollagen gene promoter luciferase reporter plasmid assay, hydroxyproline assay, gelatin zymography assay, qRT-PCR, ELISA an… Show more

“…However, many in vitro studies have indicated that ANE increases collagen formation via the transdifferentiation of myofibroblasts that express the intracellular marker protein alpha smooth muscle actin (α-SMA) and further decrease collagen degradation via the modulation of matrix metalloproteinase (MMP) activity [1315]. Furthermore, ANE may induce fibrosis through transforming growth factor-beta (TGF-β), connective tissue growth factor (CTGF), interleukin 6 (IL-6), and prostaglandin E2 (PGE2) signaling factors in different cell lines.…”

“…Tanshinone IIA is a diterpene extracted from Salvia miltiorrhiza , a popular herb that has been safely and widely used in China and other Asian countries. Previous studies have demonstrated that Tanshinone IIA exerts prominent antifibrotic effect through modulating TGF- β signaling pathway and anti-inflammatory effect through inhibiting MCP-1 expression [ 16 24 ]. Thus, we hypothesized that Tanshinone IIA might attenuate renal fibrosis after AKI via regulating recruitment of fibrocytes into the kidney through its inhibition on TGF- β and MCP-1 in a mouse model of folic acid induced kidney injury.…”

“…The latest studies in different fields have suggested the anti-fibrotic activity of Sal B in multiple organ systems such as pulmonary fibrosis, hepatic fibrosis and oral submucous fibrosis [1417, 2628]. These studies revealed the ability of Sal B to prevent fibrotic changes both in vitro and in vivo.…”
