Inhibition of Rat Intestinal Cl– Secretion by 4,5-Dimethylresorcinol

Abstract: Wood creosote, a mixture of phenolic compounds, inhibits enterotoxin-induced intestinal fluid secretion, suggesting that one of its constituents suppresses transepithelial Cl secretion from the intestinal mucosa. To identify an active constituent in wood creosote that inhibits intestinal Cl secretion through Cl channels, we first examined its effect on Cl secretion using a cultured cell line transfected with complementary DNA encoding a Clchannel and … Show more

“…Simple phenolics are some of the major components of wood creosote (Kuge et al, 2003;Morino et al, 2004;Ogata and Shibata, 2004). 4,5-Dimethylresorcinol has been shown to inhibit Cl − secretion (Ogata and Shibata, 2004). likelihood of interactions taking place.…”

“…In some countries WC, under the trade name of Seirogan, has long been used as an antidiarrheal medicine [2]. WC and its constituent compounds are known to suppress enterotoxin-induced intestinal fluid secretion via chloride channels [3][4][5] and to suppress smooth muscle contraction [6,7]. Indeed, one of the major constituents of WC, 4-ethylguaiacol, is known to possess the latter activity [8].…”

“…Moreover, wood creosote has been reported to directly inhibit the Cl − channel, cystic fibrosis transmembrane conductance regulator (CFTR) on the apical membrane. The inhibition of the Cl − channel induces an osmolality change, which suppresses water leakage [5]. Because Cl − channel hyper-activity is associated with different types of diarrhea [6], wood creosote may be effective against a wide range of diarrheal symptoms.…”

“…It is a colorless or light-yellow liquid with a peculiar smell. Although wood creosote shows only a weak antibacterial effect, it shows no or very few adverse effects [4,5]. Therefore, wood creosote has been approved as a non-prescription drug.…”
