Inhibition of Phosphodiesterase 5 and Increasing the Level of Cyclic Guanosine 3′,5′ Monophosphate by Hydroalcoholic Achillea wilhelmsii C. Koch Extract in Human Breast Cancer Cell Lines MCF-7 and MDA-Mb-468

Abstract: This study aimed to investigate the effect of hydroalcoholic Achillea wilhelmsii C. Koch extract (HAWE) on phosphodiesterase 5 (PDE5) gene expression and cyclic guanosine 3′,5′ monophosphate (cGMP) signaling in the MCF-7 and MDA-Mb-468 cell lines. The effective dose (ED50) of HAWE was examined in both cell lines using a 3-(4,5-dimethylhiazol-2-yl)-2,5-diphenyltetrazolium bromide viability test, and the type of cell death was detected by flow cytometry. The expression of PDE5 and the concentration of cGMP were … Show more

“…In recent findings, indole derivatives have the potency to treat mammary gland cancer and Tadalafil also have the indole moiety in its structure, which ultimately reveals its molecular mechanism [9]. It was reported that the expression of PDE-5 was significantly high in the malignant breast tumors as compare with normal breast tissues and benign tumors [10]. Consistently increased PDE-5 expression has also been reported in a variety of mammary gland cancer cell lines (MCF-7, T47D, HTB-26, MDA-MB-231), which gives the rationale to assess the anticancer effects of PDE-5 inhibitors in carcinogen induced animal model and check the involvement of reactive oxygen species (ROS), death receptor and mitochondrial signalling as part of the combinatorial apoptosis mechanism.…”

“…The collected leaves of P. eldarica were placed in a darkroom for drying process. Then, they were powdered and fruit extraction was obtained by Clevenger methods as described previously (12,13). Briefly, one percent of fruit powder was added to 1,000 mL of distilled water (hot) for half an hour and filtered by Whatman filter papers (11 µm pore size, No.…”

“…GC enzymes can be found in 2 major forms: soluble or cytosol form (sGC) and particulate or transmembrane form (pGC) (1). The level of this second messenger is regulated through catalysis of cGMP degradation into its inactive form (5'-GMP) by 8 phosphodiesterases (PDEs) including: dual-specificity PDEs (PDE1, PDE2, PDE3, PDE10, and PDE11) hydrolyzing both cGMP and cyclic adenosine 3', 5' monophosphate (cAMP); and cGMP-specific PDEs (PDE5, PDE6, and PDE9) only hydrolyzing cGMP (2,3).…”
