Inhibition of miR-126 protects chondrocytes from IL-1β induced inflammation via upregulation of Bcl-2

Abstract: ObjectivesThe aim of this study was to investigate the role of miR-126 in the development of osteoarthritis, as well as the potential molecular mechanisms involved, in order to provide a theoretical basis for osteoarthritis treatment and a novel perspective for clinical therapy.MethodsHuman chondrocyte cell line CHON-001 was administrated by different doses of interleukin (IL)-1β to simulate inflammation. Cell viability, migration, apoptosis, IL-6, IL-8, and tumour necrosis factor (TNF)-α expression, as well a… Show more

“…TNF-α and IL-1β have been used to trigger inflammation in chondrocyte and synoviocyte culture. Upon stimulation, the cells release IL-6 [26], IL-8 [27], IL-10 [28], IL-1β [29], and TNF-α [28]. Similar cytokine profile was increased in animal models of osteoarthritis [18,[30][31][32][33][34].…”

“…Laboratory studies showed that miR-146a [29] and miR-126 [27] were upregulated in chondrocytes induced with inflammation with lipopolysaccharide (LPS) and IL-1β, respectively. miR-146a targets CXCR4 and downregulated its expression.…”

“…Previous studies determined that chondrocyte survival was associated with joint destruction in OA patients [21,22]. The human chondrocyte cell line CHON-001, derived from normal human articular cartilage, has been widely used in vitro in osteoarthritis models [23][24][25]. Although researches have been conducted on different aspects of OA, further studies are required to be carried out on the molecular mechanisms involved in chondrocyte survival in OA.…”

“…MiR‐10a‐5p has been reported that upregulated and induced apoptosis through repression of HOXA1 in chondrocytes (Ma et al, ). In IL‐1β‐induced cell line CHON‐001, expression of IL‐6, IL‐8, and tumour necrosis factor‐α (TNF‐α) were all increased as well as miR‐126 was upregulated, while inhibition of mir‐126 reduced apoptosis and upregulated bcl‐2 expression through inactivating MAKP/JNK signaling pathway (Yu et al, ). The expression of miR‐181a‐5p was negatively correlated with selenocysteine insertion sequence elenent (SECIS)‐binding protein 2 (SBP2), GPX1 and GPX4 mRNA expression and SBP2 protein expression in the damaged cartilage were lower than normal cartilage, whereas miR‐181a‐5p expression on the contrary (Xue et al, ).…”
