Inhibition of Staphylococcus epidermidis Biofilm Formation by Rabbit Polyclonal Antibodies against the SesC Protein

Abstract: Several well-studied proteins with defined roles in Staphylococcus epidermidis biofilm formation are LPXTG motif-containing proteins. Here, we investigate the possible use of the LPXTG motif-containing protein SesC (S. epidermidis surface protein C; accession no. NP_765787) as a target for antibodies to prevent biofilm formation. In vitro and in a in vivo rat model of catheter infection, gene and protein expression analysis showed that SesC is expressed more strongly in biofilm-associated cells than in plankto… Show more

“…The fluorescent reporter was then used to compare PhnD antibodies to those for other targets, including several proteins with a known role in S. epidermidis biofilm formation: EmbP (SERP1011), Aap (SERP2398), and SesC (SERP2264) (17)(18)(19)(20). Treatment with EmbP antibodies also led to a large decrease in biofilm formation (91% reduction versus the control); however, none of the other antibodies showed substantial effects at the endpoint (Fig.…”

“…Previous investigators have utilized the indwelling catheter model in either mice or rats for investigation of genes important for biofilm formation; [31][32][33] however, it is believed that this is the first description of active vaccination with evaluation of efficacy against early biofilm formation, in this model. A relatively early time point of 24 h was chosen for efficacy evaluation, due to the kinetics of bacterial spread and/or lethality in this model.…”

“…Pure plasmid DNA was isolated using a High Pure plasmid isolation kit (Roche Diagnostics), and PCR amplification and sequencing were used for verification. The recombinant plasmids were used for recombinant protein production as previously described in detail (28). Preparation and purification of polyclonal anti-rSes IgG antibodies.…”
