Inhibition of Endotoxin-Induced Uveitis by Methylprednisolone Acetate Nanosuspension in Rabbits

Abstract: Based on our findings, we suggest that the copolymer nanosuspension may favor the localized, controlled ocular delivery of MPA for the prevention of inflammatory symptoms in ocular diseases.

“…3(A) and (B)). The X-ray spectra of the physical mixtures exposed that the reduction in the intensity of diffraction peaks was due to dilution effect exerted by the polymer network [19,20]. Concisely, the DSC and PXRD studies verified decrease of drug crystallinity in the nanoparticles.…”

“…For example, PLGA colloidal nanoparticles were exploited to deliver gene-based therapeutics to the retinal pigment epithelial cells [104] . The sustained-release nanosuspension of piroxicam and methylprednisolone acetate were formulated using Eudragit to control the endotoxininduced uveitis in rabbits [105,106] . For the treatment of chronic ocular diseases (e.g., CMV retinitis), localized prolonged nanomedicines can be used effectively as a safer alternative to the frequent injections that may cause cataract development, retinal detachment, endophthalmitis and vitreous hemorrhage [100] .…”

“…Polymeric nanosuspensions, prepared from Eudragit RL100 and RS100, have been investigated extensively for the ocular delivery of ibuprofen (18), flurbiprofen (19), cloriocromene (20), piroxicam (21), methyl prednisolone (22), and amphotericin B (23).They are cationic copolymers of methacrylate with 4-12% quaternary ammonium groups. They are inert polymer resins, insoluble at physiologic pH but have swelling properties.…”
