Inhibition of DNA synthesis in SV3T3 cultures by isolated 3T3 plasma membranes.

Abstract: 3T3 plasma membranes were added to subconfluent cultures of SV3T3 cells in the presence of fusogens. If this protocol results in the introduction into the SV3T3 cell membrane of 3T3 plasma membrane components responsible for density-dependent inhibition of growth, then the SV3T3 cell cultures would be expected to show decreased rates of DNA synthesis as they approach confluence. Results of these experiments indicate that rates of DNA synthesis in SV3T3 cultures so treated were as much as 63% less than in untre… Show more

“…Although PI 3-kinase/Akt signaling cascade is markedly increased, when PTEN is depleted, other cell signalings that are dictated by cell/cell contact remain intact in PTEN-null cells. For instance, contact inhibition can be triggered by the addition of cell membrane preparations to dividing cells in vitro (17, 18). Adhesion molecules, including particular cadherins and integrins, induce cell cycle arrest upon contact with specific components of the extracellular matrix or with neighboring cells (19, 20).…”

“…VSM cell cultures were derived from bovine aortas as described previously (Chamley et al, 1977). Swiss 3T3 fibroblasts were grown and maintained as described pre-viously (Peterson and Lerch, 1983). Cultures were grown in a 5% carbon dioxide/95% air atmosphere at 37°C in Dulbecco's Modification of Eagle's Medium (DMEM, Flow Laboratories) supplemented with penicillin, streptomycin, neomycin (100 Uiml, 100 U/ml and 100 pg/ml, respectively, Flow Laboratories), and 10% fetal calf serum (Hyclone).…”

“…Contact inhibition can be triggered by the addition of cell membrane preparations to dividing cells in vitro (e.g., Whittenberger and Glaser 1977;Peterson and Lerch 1983). A glycoprotein from such membranes could mimic the effect of cell-cell contact (Wieser et al 1990).…”
