Inhibition of Aquaporin-3 Water Channel in the Colon Induces Diarrhea

Abstract: Aquaporin (AQP) 3, which is predominantly expressed in the colon, is considered to play an important role in regulating the fecal water content in the colon. In this study, the role of AQP3 in the colon was examined using HgCl(2) and CuSO(4), which are known to inhibit AQP3 function. The fecal water content was measured up to 1 h after the rectal administration of HgCl(2) or CuSO(4) to rats. The results showed that the fecal water content in the HgCl(2) administration group increased significantly to approxima… Show more

“…AQP3 is considered to play an important role in regulating the colonic water movement. Previously, it has been demonstrated that the HgCl 2 rectal instillation affected AQP3 functioning in the colon and caused diarrhea, as fecal water content increased in mercurytreated rats about four times in comparison with controls (Ikarashi et al, 2012). In addition to the proper function of AQP3, some studies revealed a new and important function in enterocyte proliferation.…”

“…Detailed procedures have been previously described [12]. Briefly, fecal samples from the mice were collected for up to 5 h after the administration of the MgSO 4 solutions and were placed in silica gel followed by drying for 48 h in a desiccator.…”

“…Several studies have displayed the importance of AQPs in faecal concentration/water absorption in the colon. In a study on rats the inhibition of the AQP3 was shown to cause watery diarrhoea [201] and during exposure to mercury, the common inhibitor of AQPs, both AQP3 and 4 as well as 7 were downregulated and therefore believed to be involved in the mechanism of mercury-induced toxicity and gastrointestinal symptoms [202]. Wang et al also showed that AQP4 knockout mice demonstrated a higher water content in faeces [203].…”
