Inhibition of Allergic Bronchoconstriction in Guinea Pigs and in Asthmatics by the Leukotriene Antagonist ONO-1078

Abstract: The effect of the leukotriene receptor antagonist ONO-1078 on experimental and clinical bronchial asthma was evaluated. It was found that this compound could inhibit antigen-induced bronchoconstriction in passively sensitized guinea pigs and leukotriene D4- and allergen-induced bronchial responses in normal and in asthmatic subjects. Clinical trials in patients with chronic bronchial asthma demonstrated its effectiveness in reducing hazardous symptoms like wheezing and dyspneic attacks.

“…Currently, glucocorticoids, β2 agonists and leukotriene modulators are mostly used in the treatment of AA to provide rapid relief of asthma exacerbations, reduce bronchoconstriction, and decrease airway inflammation. 2,3 However, there are still some patients with unsatisfactory treatment results and long-term use of such drugs can produce significant toxic side effects. Traditional Chinese medicine has been studied extensively in the treatment of allergic diseases such as asthma, and has definite clinical efficacy, especially in reducing the side effects of western drugs, and long-term use does not reduce immunity.…”

“…Elevated levels of these mediators have been associated with the key features of the disease including, increased vascular permeability, airway obstruction, mucus hypersecretion, and leukocytic infiltration of pulmonary tissue . The improvement of acute airway function in asthmatic patients upon administration of either a peptidoleukotriene antagonist or a 5-LO inhibitor is strong evidence supporting the importance of peptidoleukotrienes in this disease . Likewise, the biochemical pharmacology of inflammatory bowel disease (IBD) suggests an important role for 5-LO products in the pathophysiology of this group of diseases.…”
