Effect of Porcine Follicular Fluid on LH and FSH Secretion in Rats . Tohoku J. exp. Med., 1988, 156 (2), 157-163 -To investigate a mode and a site of action of inhibin on FSH and LH secretion , in vivo and in vitro effects of porcine follicular fluid (PFF) on FSH and LH secretion were studied in male rats . PFF administerd i.v. suppressed basal FSH levels for 1-8 hr , but did not alter LH levels. O n the contrary, PFF given either 1 or 6 hr in advance , suppressed LH-RH stimulated LH responses as well as LH-RH induced FSH responses . In cultured pituitary cells PFF exerted a partial suppressive effect on LH release. PFF suppressed LH release only under the maximal stimulation with 10-70 M LH-RH and 0.1 mM 3-isobutyl-1-metylxanthine (IBMX) to a lesser degree compared with uniform and greater suppression on FSH release regardless of the experimental conditions. Above results showed a selective suppressive effect of PFF on FSH release and synthesis with a less inhibitory effect on LH release mainly at the pituitary level.inhibin ; porcine follicular fluid ; FSH ; LH ; LH-RH It has been widely demonstrated that follicular or testicular preparations with definite inhibin activities suppressed FSH secretion in rats (Marder et al . 1977;Schwartz and Charming 1977) as well as in other species (Channing et al. 1981 ;Rettori et al. 1982). Administration of steroid-free bovine follicular fluid (bFF), containing inhibin-like activity, depressed levels of FSH measured 4 h after injection in intact adult and 35-day-old female rats, but not in younger females. Suppression of FSH was also observed in intact male rats, aged 55 days, but not in older and younger male rats. Eight hours after injection of bFF, FSH levels were depressed in 15-day-old and older immature and adult rats of both sexes. Male and female rats, gonadectomized 2 days earlier, responded similarly to bFF treatment as did the intact animals. In a second experiment it was found that the rise of FSH levels, occurring within 8 h of gonadectomy, decreased with age in male and increased with age in female rats. Steroid treatment was found to prevent the rise in FSH levels partially in 15-day-old male and completely 25-day-old female rats, whereas treatment with bFF was fully effective in blocking the FSH rise in both immature and adult rats of both sexes. It is concluded that inhibin might be a major physiological factor in a fast-acting control of FSH concentrations from at least the age of 25 days onwards in female rats. In male rats its physiological significance might be limited to the prepubertal period, despite the fact that pituitary secretion of FSH is suppressed by exogenous inhibin-like activity at all ages studied.

A newly discovered small peptide purified from rat follicular fluid stimulates the pituitary to release FSH and LH in vitro as well as in vivo. Dialysates of crude acid extracts of ovarian follicular tissue and fluid from rats pretreated with PMS gonadotropin stimulate the secretion of both LH and FSH, but not PRL, GH, or TSH, in a pituitary monolayer culture system. This stimulating factor, named gonadocrinin for operational facility, is smaller than 3500 daltons; its biological activity disappears after treatment with trypsin. Gonadocrinin is not recognized by two-antisera binding the decapeptide LRF even though D-Phe2,D-Trp6-LR, an LRF analog antagonist, competitively inhibits the activity of ovarian gonadocrinin. Cultured rat granulosa cells also secret substances with gonadocrinin activity in vitro, indicating that the granulosa cells probably are in vivo the source of gonadocrinin. A crude preparation of gonadocrinin given iv to rats on the second day of diestrus induced secretion of LH comparable to that produced by a 250-ng LRF injection. Gonadocrinin has chemical characteristics different from those of LRF. When purified gonadocrinin or LRF was applied to an identical isocratic high pressure liquid chromatography system, LRF was eluted at a position different from that of gonadocrinin, indicating that, chemically, gonadocrinin is not identical to the hypothalamic decapeptide, LRF.
