Inhibin From Cultures of Rat Seminiferous Tubules

Abstract: Medium from cultures of mature rat seminiferous tubules contained a substance which suppressed, in a dose-related manner, the luteinizing hormone releasing hormone (LH-RH)-stimulated secretion of FSH by cultured rat pituitary cells. The secretion of LH was suppressed to a lesser extent and the basal secretion of both LH and FSH was inconsistently affected. Gel filtration on Sephadex G-100 did not fractionate the activity. The active material did not inhibit the secretion of TSH or destroy LH-RH and the activit… Show more

“…In the male, in vitro studies have shown that Sertoli cells (2)(3)(4)(5) secrete inhibin under the influence of FSH but not luteinizing hormone (LH) (5)(6)(7)(8)(9). 'The effect of LH on testicular inhibin secretion in vivo is unknown.…”

“…Early studies from this laboratory suggested that the levels of FSH are reduced without any sup¬ pression of the levels of LH when material from bovine testis or ovine rete testis fluid (RTF) is infused into wethers (male sheep castrated prepubertally), but more recent studies clearly show that both gonadotrophins are affected Blanc, Cahoreau, Courot, Dacheux, Hochereau-de-Reviers & Pisselet, 1978;Hudson et al 1979). With pituitary cell cultures there is agreement between different laboratories that inhibin reduces the gonadotrophin releasing hormone (Gn-RH)-stimulated secretion of both gonado¬ trophins: the secretion of FSH being affected more than that of LH (Baker et al 1976;Eddie, Baker, Dulmanis, Higginson & Hudson, 1978;Franchimont et al 1978;de Jong et al 1978;Labrie, Lagacé, Ferland, Kelley, Drouin, Masicotte, Bonne, Raynaud & Dorrington, 1978). However, there are several reports that the basal secretion of LH is unaffected by doses of inhibin which produce significant suppression of the basal secretion of FSH (Steinberger & Steinberger, 1976;de Jong et al 1978;Labrie et al 1978).…”
