“…Early studies from this laboratory suggested that the levels of FSH are reduced without any sup¬ pression of the levels of LH when material from bovine testis or ovine rete testis fluid (RTF) is infused into wethers (male sheep castrated prepubertally), but more recent studies clearly show that both gonadotrophins are affected Blanc, Cahoreau, Courot, Dacheux, Hochereau-de-Reviers & Pisselet, 1978;Hudson et al 1979). With pituitary cell cultures there is agreement between different laboratories that inhibin reduces the gonadotrophin releasing hormone (Gn-RH)-stimulated secretion of both gonado¬ trophins: the secretion of FSH being affected more than that of LH (Baker et al 1976;Eddie, Baker, Dulmanis, Higginson & Hudson, 1978;Franchimont et al 1978;de Jong et al 1978;Labrie, Lagacé, Ferland, Kelley, Drouin, Masicotte, Bonne, Raynaud & Dorrington, 1978). A bioassay for inhibin based on the suppression of gonadotrophin releasing hormone (Gn-RH)-stimulated secretion of FSH by primary monolayer cultures of rat anterior pituitary cells is described. The cultures were exposed to standard or test materials for 3 days. The levels of FSH in the media were measured by radioimmunoassay after exposure for 6 h to a maximally stimulating concentration of Gn-RH (10 nmol/l). The standard was prepared from ovine testicular lymph. Several preparations of proteins from gonadal tissues or secretions suppressed the levels of FSH in parallel with the standard. The levels of LH were also reduced but higher doses of active material were required. Non-specificity from cell damage and inactivation of Gn-RH have been excluded. The secretion of gonadotrophins by the pituitary cells was also inhibited by androgens, but not in parallel with the standard and secretion of LH was affected more than that of FSH. Control lymph protein preparations from castrated sheep had no detectable activity. The assay was sensitive and had adequate precision and practicability. It has proved useful for monitoring preliminary steps in the purification of inhibin.

Rat ovarian-cell culture medium contains steroids and inhibin material that are able to influence basal and LHRH-stimulated secretion of FSH and LH from dispersed pituitary cells. Studies on procedures to remove steroids, such as ethyl acetate extraction and charcoal: dextran adsorption, showed that adsorption of the culture medium with 0.5 to 1.0% charcoal and 0.05 to 0.1% dextran, respectively, was the optimal method to eliminate most of the steroids in the medium without a detrimental effect on inhibin activity. The inclusion of serum or bovine albumin in the culture medium after the culture protected the inhibin from adsorption by the charcoal: dextran treatment. The inhibin present in the rat ovarian-cell culture medium treated with 1.0% charcoal: 0.1% dextran behaved like the inhibin reference standard (from rete testis fluid of the sheep: RTFS) in the basal secretion of FSH and the LHRH-stimulated secretion of FSH and LH in the in-vitro bioassay.
