Increasing the stearic acid content to improve soybean [Glycine max (L) Merr] oil quality is a desirable breeding objective for food-processing applications. Although a saturated fatty acid, stearic acid has been shown to reduce total levels of blood cholesterol and offers the potential for the production of solid fat products (such as margarine) without hydrogenation. This would result in the reduction of the level of trans fat in food products and alleviate some current health concerns. A segregating F2 population was developed from the cross between Dare, a normal stearic acid content cultivar, and FAM94-41, a high stearic acid content line. This population was used to assess linkage between the Fas locus and simple sequence repeat (SSR) markers. Three SSR markers, Satt070, Satt474, and Satt556, were identified to be associated with stearic acid (P < 0.0001, r 2 > 0.61). A linkage map consisting of the three SSR markers and the Fas locus was then constructed in map order, Fas, Satt070, Satt474, and Satt556, with a LOD score of 3.0. Identification of these markers may be useful in molecular marker-assisted breeding programs targeting modifications in soybean fatty acids.

Soybean (Glycine max) seeds store significant amounts of their biomass as protein, levels of which reflect the carbon and nitrogen received by the developing embryo. The relationship between carbon and nitrogen supply during filling and seed composition was examined through a series of embryo-culturing experiments. Three distinct ratios of carbon to nitrogen supply were further explored through metabolic flux analysis. Labeling experiments utilizing [U- ]asparagine were used to monitor differences in carbon allocation. The analyses revealed that: (1) protein concentration as a percentage of total soybean embryo biomass coincided with the carbon-to-nitrogen ratio; (2) altered nitrogen supply did not dramatically impact relative amino acid or storage protein subunit profiles; and (3) glutamine supply contributed 10% to 23% of the carbon for biomass production, including 9% to 19% of carbon to fatty acid biosynthesis and 32% to 46% of carbon to amino acids. Seed metabolism accommodated different levels of protein biosynthesis while maintaining a consistent rate of dry weight accumulation. Flux through ATP-citrate lyase, combined with malic enzyme activity, contributed significantly to acetyl-coenzyme A production. These fluxes changed with plastidic pyruvate kinase to maintain a supply of pyruvate for amino and fatty acids. The flux maps were independently validated by nitrogen balancing and highlight the robustness of primary metabolism.

To identify genetic variation for fatty acid composition in mature soybean seeds, 4566 M 3 generation seed samples from a chemically mutagenized population were subjected to fatty acid profiling by gas chromatography. In the population, a wide range of variation in the content for each of the five major fatty acids was observed. Seventy-nine lines were identified which contained significantly high or low levels of one of the five major soybean fatty acids. These lines were advanced to the subsequent generation. Of the 79 lines showing a variant fatty acid profile in the M 3 , 52 showed clear heritability for the oil composition in the seeds of the subsequent generation. These lines are likely to represent 52 distinct genetic mutations. These mutants may represent new loci involved in the determination of soybean seed oil content or could be new isolates or alleles of previously identified genetic variants for soybean oil composition.
