Ingenol mebutate for the treatment of actinic keratosis: effectiveness and safety in 246 patients treated in real-life clinical practice

Abstract: A total of 260 treatments in 246 patients with a mean (SD) age 70.6 (10.4) years were reviewed. The number of clinically visible AK in the treated area decreased from 6.16 (3.02) to 1.22 (2.02) (p < .001) lesions with an average reduction of 84%. Univariate analysis showed higher reduction rates when IMG was applied in the face/scalp (p = .026), in women (p = .041), and in patients under 70 years of age (p = .033). According to multivariate analysis, advanced age was associated with worse clearance rates (p = … Show more

“…Moreover, although DL-PDT generally does not cause severe LSRs, in a study by Galvão et al [40], moderate erythema after two hours of outdoor exposure was seen only in an albino patient. Other proposed risk factors for greater severity of LSRs with IngMeb are: the female gender, an age below 70 years, and Korean patients [41]. This last observation was suggested to be related to the difference of race and skin thickness [42].…”

“…Some naturally occurring ingenol conjugates possess anticancer and anti-HIV properties (Abreu et al, 2014;Vasas et al, 2012), whereas semi-synthetic derivatives show potential for increased efficacy (Silva et al, 2019). Ingenol mebutate, a naturally occurring ester derivative containing angelic acid, has gained significant attention due to its approval by the Food and Drug Administration in 2012 and the European Medicines Agency in 2013 for treatment of the precancerous skin condition, actinic keratosis (Braun et al, 2018;Martin and Swanson, 2013;Ogbourne and Parsons, 2014;Ortega Del Olmo and Salido-Vallejo, 2018). Originally isolated from Euphorbia peplus, ingenol mebutate has been extensively studied for its potent antitumor and antileukemic activities (Alchin, 2014;Doan et al, 2012).…”
