“…DNA extraction, PCR amplification and sequencing DNA was extracted from mycelia of agar cultures or from dried apothecia taken from herbarium specimens. DNA was extracted and amplified using PCR following the methods of Peláez et al (1996) or Johnston and Park (2013). Amplification primers used for the ITS1-5.8S-ITS2 region were ITS1F, ITS5, ITS4, or ITS4a (White et al 1990;Gardes and Bruns 1993;Larena et al 1999), for the LSU region were LROR and LR5 (Bunyard et al 1994;Vilgalys and Hester 1990), and for the SSU region were NS1 and NS4 (White et al 1990).…”

“…DNA extraction, PCR amplification, and DNA sequencing DNA was extracted from small basidiomata fragments of Steccherinum lusitanicum AH 31879 and Mycoacia nothofagi AH 31887 following previously described methods (Peláez et al 1996). Also, three culture strains of Phlebia setulosa (FP 106520, HHB 11749, RLG 5105), kindly submitted by K. Nakasone (Center for Forest Mycology Research, Madison Field Office, Wisconsin USA), were also subjected to DNA isolation for sequencing.…”

“…Investigating the diversity of functional genes is more advanced in bacteriology, where prototype gene arrays are available (e.g., Wu et al 2001). Intraspecific diversity of aquatic hyphomycetes was first studied by Pelá ez et al (1996) with the RAPD technique (random amplified polymorphic DNA) applied to the 5.8S rRNA of strains of Heliscus lugdunensis and Articulospora tetracladia, both with known teleomorphs. They found 7 and 5 RAPD types in the two species, randomly distributed in foam patches along a 1 km long stream reach.…”
