Infrared Spectroscopy with Multivariate Analysis Potentially Facilitates the Segregation of Different Types of Prostate Cell

Abstract: The prostate gland is conventionally divided into zones or regions. This morphology is of clinical significance as prostate cancer (CaP) occurs mainly in the peripheral zone (PZ). We obtained tissue sets consisting of paraffin-embedded blocks of cancer-free transition zone (TZ) and PZ and adjacent CaP from patients (n = 6) who had undergone radical retropubic prostatectomy; a seventh tissue set of snap-frozen PZ and TZ was obtained from a CaP-free gland removed after radical cystoprostatectomy. Paraffin-embedd… Show more

“…Principal Components Analysis (PCA) is a method of multivariate analysis broadly used with datasets of multiple dimensions (Varmuza, 2009;Korenius et al, 2007;German et al, 2006). It allows the reduction of the number of variables in a multidimensional dataset, although it retains most of the variation within the dataset.…”

“…Thus, to accurately model its role, we used albumin and chymotrypsin to represent the ␣-helix and ␤-sheet conformations and found that a 50:50 mixture best fit the amide I and II peaks. Although other calibration models such as principal component analysis (29) and neural networks (30) may be better suited to analyze the fine structure of the FTIR spectra, a linear model provides an explanation of the sources of infrared absorption in terms of physiological parameters.…”

“…For instance, Matthew German and colleagues [68] coupled high-resolution synchrotron radiation-based FTIR (SR-FTIR) microspectroscopy with PCA to investigate the characteristics of putative adult stem cell (SC), transiently amplified (TA) cell, and terminally differentiated (TD) cell populations of the corneal epithelium. Using PCA, each spectrum, composed by many variables (the wavenumbers), is reduced to a point in a low dimensional space.…”
