Influenza Viruses: Innate Immunity and mRNA Vaccines

Abstract: The innate immune system represents the first line of defense against influenza viruses, which cause severe inflammation of the respiratory tract and are responsible for more than 650,000 deaths annually worldwide. mRNA vaccines are promising alternatives to traditional vaccine approaches due to their safe dosing, low-cost manufacturing, rapid development capability, and high efficacy. In this review, we provide our current understanding of the innate immune response that uses pattern recognition receptors to … Show more

“…Recently developed mRNA-based vaccines have demonstrated great promise after proven efficacious against severe COVID-19 outcomes, including hospitalization and death. HA based mRNA vaccines are in clinical trials, as reviewed in ( 230 ) and may have some advantages over conventional inactivated influenza vaccines ( 231 233 ). Another approach is to develop a next generation influenza vaccine which triggers heterosubtypic or heterologous immunity ( 234 ).…”

“…Inflammasome sensors can recognize PAMPs and/or DAMPs produced during pathogen infection and cellular instability, respectively (Kanneganti, 2010). Among the sensors, the most wellcharacterized sensor is the NLR family pyrin domaincontaining 3 (NLRP3), which has been implicated in several diseases such as autoinflammatory diseases, obesity, colitis, and pathogen-mediated diseases including influenza A viruses and corona viruses (Gurung & Kanneganti, 2016;Kanneganti et al, 2006aKanneganti et al, , 2006bKarki et al, 2015;Lee et al, 2020;Lee & Ryu, 2021;Stienstra et al, 2011;Thomas et al, 2009;Zaki et al, 2010).…”
