Influence of Warm Ischemia Time on Peripheral-Type Benzodiazepine Receptor: A New Aspect of the Role of Mitochondria

Abstract: The peripheral benzodiazepine receptor (PBR) is located mainly in the outer mitochondrial membrane and many functions are associated directly or indirectly with the PBR. We have studied the influence of different durations of warm ischemia (WI) on renal function, tissue damage and PBR expression in a Large Whitepig model. After a midline incision, the renal pedicle was clamped for 10 (WI10), 30 (WI30), 45 (WI45), 60 (WI60) or 90 min (WI90), and blood and renal tissue samples were collected between 1 day and 2 … Show more

“…First, we demonstrated that TAC resulted in a 3-fold increase in expression of the TSPO in WT animals (Fig. 1F,G ), an upregulation of TSPO that parallels that seen in cardiac ischemia and reperfusion 29 and under stress conditions in other tissues 27 , 28 , 30 . Pressure overload for 8 weeks in WT animals resulted in the expected changes in cardiac morphology, function, and metabolism associated with HF (see Figs 2 and 3 , and Table 1 ).…”

“…The pig seems to be the ideal species to serve this purpose. 1 Several porcine models to study mechanisms of WI, CI and IRI exist: reperfusion models 2,3 , isolated organ preservation systems 4,5 and kidney transplantation models. [6][7][8][9] However, the technical and surgical aspects of these models and their possible pitfalls have not been described in detail.…”
