Influence of the ABC Transporter YtrBCDEF of Bacillus subtilis on Competence, Biofilm Formation and Cell Wall Thickness

Abstract: Bacillus subtilis develops genetic competence for the uptake of foreign DNA when cells enter stationary phase and a high cell density is reached. These signals are integrated by the competence transcription factor ComK, which is subject to transcriptional, post-transcriptional and post-translational regulation. Many proteins are involved in the development of competence, both to control ComK activity and to mediate DNA uptake. However, for many proteins, the precise function they play in competence development… Show more

“…Cell wall modification is one of several mechanisms of being involved in antibiotic resistance and has been shown to play an important role in the resistance mechanism in S. aureus , C. difficile , S. pneumoniae and S. agalactiae 48 50 . Also, in recent studies it is suggested that transporters can have a direct or indirect influence on peptidoglycan biosynthesis or peptidoglycan remodeling as was shown recently for the ABC transporter YtrBCDEF in B. subtilis 51 .…”

“…Genes involved in these pathways have also been observed in other transcriptomic and experimental studies. IVM-An biofilm bacteria showed the highest expression of genes encoding different ABC transporters, which suggested that these bacteria might be less metabolically active compared to bacteria in aerobic biofilms 44 . This assumption was further supported by the lower expression of genes involved in ribosome organisation, indicative of a lower growth rate of IVM- An biofilm bacteria.…”

“…While emphasizing a low Spo0A~P as a main cause of inefficient expression of the comK gene in Rho + cells, these results strongly suggest that a stable high level of Rho also impedes the expression of other gene(s) directly or indirectly involved in the development of genetic competence. There is nothing unusual about this, since many proteins that have been shown to be essential for genetic transformation act independently and downstream of ComK [80,[114][115][116]. Interestingly, many of these proteins play essential roles in the RNA metabolism [114][115][116].…”

Interestingly, many of these proteins play essential roles in the RNA metabolism [114][115][116]. As in the case of Rho, their respective roles in the development of genetic competence remain elusive [114,116].…”
