This study was done to determine the effects of processing techniques on the quality of semen from Dutch AI-bucks with the view on improving pregnancy rates after artificial insemination (AI) with liquid or frozen-thawed semen. Motility of spermatozoa was estimated under a microscope whereas the percentage live spermatozoa and the percentage live spermatozoa with intact acrosomes were determined by means of flow cytometry. Aspects of semen processing that were investigated are storage temperature of liquid semen (i), the effect of glycerol on liquid-stored semen (ii), removal of seminal plasma (iii) and type of extender (iv). The correlation between semen quality and fertility rates in inseminated does was also investigated. openUP (July 2007)The percentage motile spermatozoa in semen stored in liquid form for 72 h progressively declined over time, irrespective of whether storage occurred at 4 or 18 °C. The percentage motile spermatozoa in semen stored at 18 °C was similar to that in semen stored at 4 °C if stored for 24 h but lower if stored for 48 h. Goats differ in the sensitivity of their spermatozoa to the deleterious effects of glycerol. Neither the removal of seminal plasma nor the type of extender had any effect on semen quality before freezing but semen frozen in a Tris-citric acid-glucose (TCG) buffer with egg yolk without removal of the seminal plasma had better quality after thawing than semen frozen in another diluent or after removal of seminal plasma.Remarkably no significant correlation between fertility and membrane integrity of spermatozoa could be found. Thus, although integrity assays for spermatozoa are useful to asses resistance to semen handling, the validity of these assays for predicting fertility is questioned.

Cryopreservation of Aceh Swamp Buffalo (Bubalus bubalis) Semen with Combination of Glycerol and Lactose

Aceh swamp buffaloes (Bubalus bubalis) are decreasing their population and genetic quality. This study was conducted to determine the influence of lactose and glycerol cryoprotectants on spermatozoa of Aceh swamp buffaloes after thawing. This study used completely a factorial randomized design with nine treatments, and five replications. Fresh semen of the Aceh swamp buffalo were diluted by using a combination extender lactose cryoprotectants 0 mM (L 0 ), 60 mM (L 60 ), 120 mM (L 120 ) and glycerol 3% (G 3 ), 5% (G 5 ), 7% (G 7 ) with the equilibration of 4 hours. The results showed that the combination of cryoprotectants L 120 G 7 influenced significantly (P < 0.05) on the quality of spermatozoa of the Aceh swamp buffalo (B. bubalis) after thawing. The percentage of sperm motility L 120 G 7 (42.60 ± 1.14); viability L 120 G 7 (55.00 ± 0.71); acrosome integrity L 120 G 7 (52.00 ± 0.71); and plasma membrane integrity L 120 G 7 (53.20 ± 1.48). The combination of lactose cryoprotectants 120 mM (L 120 ) and glycerol 7% (G 7 ) was the best combination to maintain the quality of spermatozoa of swamp buffalo. This finding could be used to define a policy for the spermatozoa storage of Aceh swamp buffalo to artificial insemination (AI).

The objective of this study was to test the use of a commercial extender (Triladyl) as a diluent in caprine semen refrigerated at 15˚C, using caffeine (CF), heparin (HP), synthetic oviductal fluid (SOF) andtriladyl (TRY) as capacitating chemical agents at different times. Twenty ejaculates of caprine semen were collected using an artificial vagina. The ejaculates were diluted and refrigerated by three days. Evaluated the progressive motility (PM) and the functional state of the sperm plasma membrane trough fluorescent CTC staining, counting 200 spermatic cells (non-capacitated spermatozoa NCS, capacitated CS and reacting spermatozoa REA) of caprine in two capacitating agents: CAF and HEP; one culture medium: SOF and a commercial extender: TRY at 60, 120, 180 and 240 min of incubation, during 24, 48 and 72 hs. PM was high under TRY, and CS was high under the HEP treatment. TRY could be an alternative to capacitate caprine spermatozoa, keeping PM for a longer time than HEP or CAF.
