Influence of Quorum Sensing and Iron on Twitching Motility and Biofilm Formation in Pseudomonas aeruginosa

Abstract: Reducing iron (Fe) levels in a defined minimal medium reduced the growth yields of planktonic and biofilm Pseudomonas aeruginosa, though biofilm biomass was affected to the greatest extent and at FeCl 3 concentrations where planktonic cell growth was not compromised. Highlighting this apparently greater need for Fe, biofilm growth yields were markedly reduced in a mutant unable to produce pyoverdine (and, so, deficient in pyoverdine-mediated Fe acquisition) at concentrations of FeCl 3 that did not adversely af… Show more

“…Expression of the lasI gene is maximal at day 4 of biofilm development, decreasing between days 6 and 8. The expression of rhlI fluctuates during biofilm development and phenotypes of biofilm development with a rhlI mutant vary, according to the media and model used, supposedly due to different iron levels present (Davies et al, 1998;De Kievit et al, 2001;Patriquin et al, 2008;Yoon et al, 2002).…”

“…Genetic determinants that play a role in biofilm formation have been unravelled in different bacterial species, and environmental and cellular signals that influence this process have also been described. These include iron availability, quorum sensing, and the intracellular secondary messenger cyclic di-GMP (Banin et al, 2006;Patriquin et al, 2008;Ueda & Wood, 2009;Coenye, 2010). Changes in the levels of cyclic di-GMP correlate with phenotypic changes associated with virulence, motility, colony morphology, production of exopolysaccharides, and the transition between planktonic and sessile lifestyles (Hengge, 2009;Römling & Simm, 2009, and references therein).…”

“…The effect of iron was confirmed with a mutant defective in pyoverdine mediated iron transport (Patriquin et al 2008, Ponraj et al 2012. The intracellular iron concentration serves as a signal for P. aeruginosa biofilm development .…”

“…Several mechanisms for a biofilm promoting effect of iron have been proposed for various species , Patriquin et al 2008), but its role in biofilm formation of B. cereus remains to be determined. The fact that both under iron excess and iron limited conditions a linear correlation was found between biofilm CFUs and CV stained biomass (Fig.…”
