Influence of Methylenetetrahydrofolate Reductase C677T, A1298C, and G80A Polymorphisms on the Survival of Pediatric Patients with Acute Lymphoblastic Leukemia

Abstract: The influence of genic polymorphisms involved in metabolism of chemotherapeutic agents as the methotrexate (MTX) has been studied mainly in acute lymphoblastic leukemia (ALL) of childhood. Advances in treatment may be attributed to identification of prognostic factors added to chemotherapy protocol. The aim of this study was to analyze the association of the C677T, A1298C, and G80A polymorphisms on MTHFR gene and on the overall survival of pediatric patients (n = 126) with lymphoblastic leukemia treated with M… Show more

“…Some studies have indicated that these variants play protective roles [32,33,38], whereas others have shown that they are linked to increased rates of relapse and drug resistance [5,6,7,8,9,15,18,23,24,25,26,27,28]. Among these gene polymorphisms, the higher enzyme activity of the TS 3R/3R variant led to the diminished MTX effect and enhanced drug resistance.…”

“…Although there are several reports showing associations between these polymorphisms and ALL risk [8], [42], [43], or survival [23], our results correspond to the results of a meta-analysis where the pooled odds ratio for these two variants did not show association with ALL risk [44], and also to some recent and relatively large studies, where neither SNPs showed association [26], [27]. In these papers it is also discussed in details what the explanations for these inconsistent findings can be.…”

“…The A1298C SNP was also associated with myelotoxicity in three studies although two studies found this polymorphism to be protective . Only a single study associated the G80A SNP with myelotoxicity . Two studies investigated the replication factor C ( RFC ) gene.…”
