Influence of Leishmania RNA Virus 1 on Proinflammatory Biomarker Expression in a Human Macrophage Model of American Tegumentary Leishmaniasis

Abstract: Compared to LRV-1-negative L. V. braziliensis, LRV-1-positive strains of L. V. braziliensis produced a predominant Th2-biased immune response, correlated in humans to poorer immunologic control of infection and more severe disease, including mucosal leishmaniasis. Effects of LRV-1 on the pathogenesis of American tegumentary leishmaniasis may be species specific.

“…Considering the observation of LRV2 in L. major , L. aethiopica and L. infantum in OWL outside of South America, these data suggest that LRV appeared before the divergence of the L. ( Leishmania ) and ( Viannia ) subgenera (Zangger et al, ). In the New World (in regions of Central and South America), in L. Viannia complex, LRV has been detected in strains of L. (Viannia) guyanensis , L. (Viannia) braziliensis , L. (Viannia) panamensis , L. (Viannia) lainsoni and L. V. amazonensis from L. mexicana complex (Cantanhêde et al, ; Kariyawasam et al, ).…”

“…Recently, the presence of LRV1 in L. (V) panamensis isolates from Ecuador and Costa Rica was reported showing that LRV1 is broadly spread into Leishmania species from the Viannia subgenus at the north of the Amazon Region (15).…”

“…At the epidemiologic level, LRV1/2 has been found at frequencies ranging from 0 to 87% in clinical samples from patients with leishmaniasis (15, 17, 1933); the strength of the association between viral presence and complication development varies according to the study (24, 28) and among different South-American Regions (30). The wide range in LRV1 frequency of detection in clinical samples may reflect diverse experimental approaches that do not account for differences in clinical specimens.…”

“…LRV-sequences were also detected in an Iranian L. infantum clinical isolate from a patient unresponsive to antimonial treatment (7). Viral coinfections and presence of Leishmania RNA virus are therefore increasingly perceived as risk factors for pathogenicity of human leishmaniasis (5,(7)(8)(9). The virus appears to use the Leishmania exosomal pathway to reach the extracellular environment (10).…”
