Influence of Lamotrigine and Topiramate on MDR1 Expression in Difficult‐to‐Treat Temporal Lobe Epilepsy

Abstract: Summary:Purpose: Overexpression of the multiple drug resistance gene 1 (MDR1) was quantified in brain tissue from Coriaria lactone (CL)-kindled Sprague-Dawley (SD) rats after treatment with lamotrigine (LTG) or topiramate (TPM) and compared with that found in rats treated with carbamazepine (CBZ) and valproate (VPA).Methods: Twenty-five CL-kindled SD rats were randomized into five groups (n = 5 for each group) to receive once-daily feeding of CBZ, VPA, TPM, and LTG as the monotherapy equivalent of maximum huma… Show more

“…For semiquantiWcation of the average optical density (OD) of cholinesterase-positive nerves and M 2 receptors, the method followed that in a previous study (Wang-Tilz et al 2006). Images of the stained sections were acquired and analyzed with Image Pro Plus 5.1, a computer-assisted image analysis system.…”

“…Immunoreactive staining was characterized quantitatively by digital image analysis using the Image Pro-Plus 6.0 (Media Cybernetics, Inc., Bethesda, Md., USA), as reported in [26] and identical to what we have published previously [21,22] without prior knowledge of any of the clinicopathological information. Briefly, images were obtained with a microscope (Olympus BX51; Olympus, Tokyo, Japan) fitted with a digital camera (Olympus DP70; Olympus).…”
