Influence of chamber type integrated with computer-assisted semen analysis (CASA) system on the results of boar semen evaluation

Abstract: The objective of the study was to evaluate the effect of different types of chambers used in computer-assisted semen analysis (CASA) on boar sperm concentration and motility parameters. CASA measurements were performed on 45 ejaculates by comparing three commonly used chambers: Leja chamber (LJ), Makler chamber (MK) and microscopic slide-coverslip (SL). Concentration results obtained with CASA were verified by manual counting on a Bürker hemocytometer (BH). No significant differences were found between the con… Show more

“…The above-mentioned researchers used a Leja chamber in their estimates, while a Makler chamber was used in the present study. Many researchers have stated that the CASA outcomes could be influenced by the analysis’ chambers [ 41 , 42 , 43 ]. In addition, in the present study, low-motility ejaculates were initially rejected because they could not be used for artificial insemination.…”

“…The SCA system is based on the analysis of 25 consecutive digitized photographic images obtained from a single field under negative phase contrast and at 10× magnification. The parameters were adjusted to boar sperm, and the particle area was set at 10–80 μm [ 22 ]. Sperm with an average path rate (VAP) equal to or higher than 10 μm/s were classified as immotile.…”

“…However, one aspect that has been evaluated recently is the type of slide chamber used during the CASA assessment. In the study by Gączarzewicz (2015) three slide types were used: Leja chamber; Makler chamber; and a common glass microscope slide with a coverslip. The most important differences among these slides were the depths of the chamber in which the sperm were loaded and analyzed.…”
