Influence of Carvedilol on Superoxide Generation and Enzyme Release From Stimulated Human Neutrophils

Abstract: Activation of neutrophils induces generation of reactive oxygen species and release of granule enzymes, which not only participate in the bactericidal mechanisms of these cells, but also in possible tissue damage. We studied the effect of carvedilol (CARV) [0.1-100µmol/l], an antihypertensive and cardiovascular drug with antioxidative properties, on superoxide generation (SO) and myeloperoxidase (MPO) release from isolated human neutrophils stimulated with fMLP, a specific receptor activator, or with PMA, a re… Show more

“…Glutathione in its reduced form is the most abundant intracellular antioxidant and is involved in the direct scavenging of free radicals or serving as a substrate for the glutathione peroxidase enzyme that catalyzes H 2 O 2 detoxification. Catalase is also known to be a protective enzyme and functions for the detoxification of highly reactive free radicals (Mačičková et al, 2005).…”

“…The oxidative stress is mediated due to free radicals, so it is necessary to account for antioxidant enzymes that serve as the first line of defence against radicals. CAT, GSH, and TBARS enzymes all account for neuron protection in oxidative stress [ 72 ]. GSH levels are known for disrupting neurons in neurodegenerative diseases [ 73 ].…”

“…Carvedilol has been reported in the literature as acts as a NO modulatory agent in vascular endothelial cells and in cell-free systems (Yoshioka et al, 2000). It has also been shown to inhibit superoxide ion release from activated neutrophils (Maèièkova et al, 2005).Carvedilol also has been shown to preserve the endogenous antioxidant system (i.e., vitamin E andreduced glutathione), which is normally consumedwhen tissues or cells are exposed to oxidative stress . This preservation of endogenous antioxidants may explain why the Carvedilol treatment was able to reverse scopolamine induced cognitive deficit in mice.…”
