Influence of Albumin Configuration by the Chiral Polymer-Grafted Gold Nanoparticles

Abstract: The interaction between nanoparticles (NPs) and proteins is a topic of high relevance for the medical application of NPs. This study reveals the molecular chirality on NP surfaces as an indirect regulator of the interaction between proteins and NPs. Poly(N-acryloyl-valine) (PAV) polymers with d- and l-configurations were conjugated onto gold NPs with a size of 5 nm to obtain the l-PAV-AuNPs and d-PAV-AuNPs, respectively. They had same chemical composition and surface grafting density but different surface chir… Show more

“…However, the adhesion of NP to the cell membrane is difficult to disentangle in the presence of simultaneous internalization. Thus, L-lecithin molecules, a type of phospholipids and composed of phosphoric acid with choline, glycerol or other fatty acids, were used as a model to study the interactions of chiral PAV molecules and lipids by isothermal titration calorimetry (ITC) 45 . As shown in Fig.…”

“…The data were analyzed to determine the binding stoichiometry ( N ), complex stability constant ( K ), standard molar reaction enthalpy (Δ H ) and other thermodynamic parameters of the reaction by using the supplied Origin 7.0 software. One set of binding sites model was used to fit the data as reported previously 45 50 . The reported thermodynamic parameters were an average of duplicate experiments.…”

“…As one example, poly( N ‐acryloyl‐valine) polymers with d ‐ and l ‐configuration were covalently grafted on GNPs to investigate polymer chirality effects on the interactions between protein and GNPs . A layer‐by‐layer polyelectrolyte coating strategy was applied for GNP surface modification with cationic polyallylamine and anionic polystyrenesulfonate, and the abundant surface‐exposed amino groups present after the final coating of a polyallylamine layer were used to covalently graft an antibody via its carboxylic acid groups of, e.g., its C‐terminal amino acid by amide coupling .…”

“…[9] However,t he interactions between optically active polymers and enzymes have not been widely explored. [10] We postulate that enzymes may show different performances towardd ynamers containing componentso fo pposite chirality.T his is of speciali nterest for dynameric enzyme activation for applications like carbon dioxides equestration.…”

“…Optically active polymers have been intensively studied during the last decades, and have been applied in chiral recognition/separation of enantiomers and self‐assembly of complex nanostructures among others . However, the interactions between optically active polymers and enzymes have not been widely explored . We postulate that enzymes may show different performances toward dynamers containing components of opposite chirality.…”
