f Feed supplementation with the probiotic Enterococcus faecium for piglets has been found to reduce pathogenic gut microorganisms. Since Escherichia coli is among the most important pathogens in pig production, we performed comprehensive analyses to gain further insight into the influence of E. faecium NCIMB 10415 on porcine intestinal E. coli. A total of 1,436 E. coli strains were isolated from three intestinal habitats (mucosa, digesta, and feces) of probiotic-supplemented and nonsupplemented (control) piglets. E. coli bacteria were characterized via pulsed-field gel electrophoresis (PFGE) for clonal analysis. The high diversity of E. coli was reflected by 168 clones. Multilocus sequence typing (MLST) was used to determine the phylogenetic backgrounds, revealing 79 sequence types (STs). Pathotypes of E. coli were further defined using multiplex PCR for virulence-associated genes. While these analyses discerned only a few significant differences in the E. coli population between the feeding groups, analyses distinguishing clones that were uniquely isolated in either the probiotic group only, the control group only, or both groups (shared group) revealed clear effects at the habitat level. Interestingly, extraintestinal pathogenic E. coli (ExPEC)-typical clones adhering to the mucosa were significantly reduced in the probiotic group. Our data show a minor influence of E. faecium on the overall population of E. coli in healthy piglets. In contrast, this probiotic has a profound effect on mucosa-adherent E. coli. This finding further substantiates a specific effect of E. faecium strain NCIMB 10415 in piglets against pathogenic E. coli in the intestine. In addition, these data question the relevance of data based on sampling fecal E. coli only.

Through these activities probiotics can support three core benefits for the host: supporting a healthy gut microbiota, a healthy digestive tract and a healthy immune system. More recently, the concept of combining probiotics and prebiotics, i.e. synbiotics, for the beneficial effect on gut health of pigs has attracted major interest, and examples of probiotic and prebiotic benefits for pigs are pathogen inhibition and immunomodulation. Yet, it remains to be defined in pigs, what exactly is a healthy gut. Because of the high level of variability in growth and feed conversion between individual pigs in commercial production systems, measuring the impact of probiotics on gut health defined by improvements in overall productivity requires large experiments. For this reason, many studies have concentrated on measuring the effects of the feed additives on proxies of gut health including many immunological measures, in more controlled experiments. With the major focus of studying the balance between gut microbiology, immunology and physiology, and the potential for prevention of intestinal disorders in pigs, we therefore performed a literature review of the immunomodulatory effects of probiotics, either alone or in combination with prebiotics, based on in vivo, in vitro and ex vivo porcine experiments. A 3 consistent number of studies showed the potential capacity in terms of immunomodulatory activities of these feed additives in pigs, but contrasting results can also be obtained from the literature.Reasons for this are not clear but could be related to differences with respect to the probiotic strain used, experimental settings, diets, initial microbiota colonization, administration route, time and frequency of administration of the probiotic strain and sampling for analysis. Hence, the use of proxy measurements of enteric health based on observable immunological parameters presents significant problems at the moment, and cannot be considered robust, reliable predictors of the probiotic activity in vivo, in relation to pig gut health. Sistema inmune. resUMenEl destete produce un periodo breve de ayuno y adaptación a una nueva ración sólida, la cual provoca disminución de la supervivencia de los lechones y proliferación de la microbiota patógena. Se ha propuesto la utilización de probióticos, evitando que los seres humanos consuman alimentados tratados con antibióticos. El objetivo de este trabajo fue determinar el efecto de la adición de cepas de probióticos en cerdos en crecimiento sobre las poblaciones celulares del sistema inmune (eosinófilos, basófilos, neutrófilos, monocitos y linfocitos) en intestino delgado. Se sacrificaron 35 lechones escalonadamente los días 1 (21 días de edad), 15 y 30 posdetete, y se extrajo completamente el intestino. Los animales fueron alimentados con dos dietas: dieta comercial con y sin la adición de antibiótico; a esta última se adicionaron los diferentes probióticos (Lactobacillus casei, Lactobacillus acidophilus o Enterococcus faecium) en el agua de bebida. Se utilizó un diseño de bloques al azar en un arreglo de parcelas divididas. Los aislamientos realizados a partir de las muestras intestinales de E. faecium presentaron un mayor número de poblaciones celulares del sistema inmune a nivel intestinal (p<0.01). Además, se realizó medición del pH intestinal, el cual presentó una disminución estadística significativa (p<0.01), donde los animales que consumieron E. faecium presentaron los valores más bajos de pH intestinal, frente a aquellos que consumieron la dieta con adición de antibiótico. La adición de la cepa E. faecium en el alimento de lechones recién destetados, estimula el aumento en el número de poblaciones celulares intestinales del sistema inmune y alcanzan una mayor disminución del pH intestinal. Weaning originates a brief period of fasting and adaptation to a new solid portion, which causes decreased survival of piglets and proliferation of pathogenic microbiota. It has proposed the use of probiotics, preventing humans consume fed treated with antibiotics. Therefore, the objective of this work was to determine the effect of the addition of probiotic strains in growing pigs on immune cell populations (eosinophils, basophils, neutrophils, monocytes and lymphocytes) in the small intestine. Thirty-five piglets stepwise days 1 (21 days old), 15 and 30 posdetete were sacrificed, and the intestine was completely removed. The animals were fed two diets: commercial diet with and without the addition of antibiotic; the latter different probiotic (Lactobacillus casei, Lactobacillus acidophilus or Enterococcus faecium) were added in the drinking water. A design randomized block was used in a split plot arrangement. Isolates obtained from the intestinal samples of E. faecium showed a greater number of immunological intestine cell populations (p<0,01).
