Synthesis and Application of Scaffolds of Chitosan-Graphene Oxide by the Freeze-Drying Method for Tissue Regeneration

Several biomaterials, including natural polymers, are used to increase cellular interactions as an effective way to treat bone injuries. Chitosan (CS) is one of the most studied biocompatible natural polymers. Graphene oxide (GO) is a carbon-based nanomaterial capable of imparting desired properties to the scaffolds. In the present study, CS and GO were used for scaffold preparation. CS was extracted from the mycelium of the fungus Aspergillus niger. On the other hand, GO was synthesized using an improved Hummers-Offemann method and was characterized by Fourier transform infrared spectroscopy (FTIR), Raman spectroscopy, atomic force microscopy (AFM), X-ray diffraction (XRD), and dynamic light scattering (DLS). Subsequently, three formulations (GO 0%, 0.5%, and 1%) were used to prepare the scaffolds by the freeze-drying technique. The scaffolds were characterized by FTIR, thermogravimetric analysis (TGA), and scanning electron microscopy (SEM), to determine their thermal stability and pore size, demonstrating that their stability increased with the increase of GO amount. Finally, the scaffolds were implanted, recollected 30 days later, and studied with an optical microscope, which evidenced the recovery of the tissue architecture and excellent biocompatibility. Hence, these results strongly suggested the inherent nature of chitosan/graphene oxide (CS/GO) scaffolds for their application in bone tissue regeneration.

Introduction/Objective The aim of this paper was to verify the biocompatibility of the newly synthesized nanostructured material based on calcium aluminate after implantation into the subcutaneous tissue of rats. Methods The study included 18 rats aged 10-11 weeks, divided into two experimental groups (n = 9). In all animals, incision took place on the back and two pockets of 15 mm in depth were made, in which sterile polyethylene tubes with test materials [calcium aluminate cement (ALBO-CA), calcium silicate cement with the addition of hydroxyapatite (ALBO-CSHA), and mineral trioxide aggregate (MTA) for the control group) were implanted. Six rats of each group were sacrificed in three observational periods (seven, 15, 30 days). Pathohistological analysis included inflammation, bleeding, fibrous capsule, and tissue integrity around the implanted material. Results After seven days of treatment, ALBO-CA and ALBO-CSHA showed better tissue response compared to MTA, with a statistically significant difference in inflammation intensity (p = 0.2781). The difference in vascular congestion and thickness of the fibrous capsule after implantation of ALBO-CA material compared to MTA was also statistically significant (p = 0.5567). At the end of the 30-day evaluation period, an identical inflammatory response of connective tissue at the site of implanting ALBO-CA, ALBO-CSHA, and MTA (score of 0.7) was recorded. The formation of thick or moderately thick fibrous capsule was found to be the thickest in ALBO-CA (grade 3.7). There were no statistically significant differences between the parameters analyzed after 30 days. Conclusion Newly synthesized ALBO-CA showed a satisfactory tissue response and confirmed biocompatibility after implantation in subcutaneous tissue of rats.

This study aimed to evaluate the Carbon Fiber obtained from PAN textile and cotton fiber in their different forms of presentation: non‐activated carbon fiber felt (NACFF), activated carbon fiber felt (ACFF), silver activated carbon fiber felt (Ag‐ACFF), and activated carbon fiber tissue (ACFT), to obtain scaffolds as a potential material with properties related to the synthetic bone graft. Characterization tests performed: surface wettability, traction, swelling, and in vivo tests: evaluation of the inflammatory response by implanting the materials in the subcutaneous tissue of 14 Wistar rats, evaluation of collagen fibers by picrosirius red staining and assessment of toxicity in the following organs: heart, spleen, liver, and kidney. In the wettability test, NACFF and ACFT were hydrophobic (θ124° and 114°), ACFF and Ag‐ACFF were hydrophilic. For maximum stress, ACFF was more resistant (2.983 ± 1.059) p < .05. In the swelling test, the Ag‐ACFF and ACFF groups showed the highest absorption percentage for the PBS solution and distilled water (p < .001). The organs showed no signs of acute systemic toxicity. The implant regions showed mild to moderate inflammatory infiltrate at 7 and 21 days. Only the ACFT group did not show the maturation of type I collagen fibers in 21 days. Through the conducted analyses, the ACFT shows little potential to be indicated as a possible scaffold. Therefore NACFF, ACFF, and Ag‐ACFF have the potential to be considered scaffolds due to the following characteristics presented: good absorption rate, hydrophilicity, and non‐toxic.
