Inflammation-Responsive Drug-Conjugated Dextran Nanoparticles Enhance Anti-Inflammatory Drug Efficacy

Abstract: Stimuli-responsive nanoparticles (NPs) are especially interesting to enhance the drug delivery specificity for biomedical applications. With the aim to achieve a highly stable and inflammation-specific drug release, we designed a reactive oxygen species (ROS)-responsive dextran-drug conjugate (Nap-Dex). By blending Nap-Dex with the acid-sensitive acetalated dextran polymer, we achieved a dual-responsive NP with high specificity toward the inflammatory environment. The inflammatory environment not only has elev… Show more

“…Paw edema induced by dextran administration is a model characterized by an increase in vascular permeability, kinin activation and mast cell degranulation with histamine and serotonin release, resulting in edema with a small quantity of neutrophils and proteins [38,54]. Once released, histamine and serotonin quickly act on blood vessels, triggering vasodilation with a consequent increase in blood flow to the injured site [38,54].…”

“…When tissue injury happened, fibrin clot was formed by fibrinogen and platelets. As for dextran and fibrin, they have been approved by the FDA and widely employed in biomedical applications, such as anti-inflammatory ( Lee et al, 2018 ; Tanaka et al, 2019 ) and tissue regeneration ( Marcinczyk et al, 2017 ) drug delivery.…”

“…Furthermore, in vivo anti-inflammatory effects of curcumin-loaded nanoparticles were validated in a mouse model with ankle inflammation induced by LPS, revealing significant mitigation of oxidative stress. Along this line, reactive oxygen species (ROS)-sensitive dextran-naproxen conjugate was blended with acid-responsive acetalated dextran polymer, forming a dually responsive nanoparticle with high specificity towards inflammatory microenvironment [ 134 ].
