Inflammation inhibitory effect of water extract from pumpkin’s tendril

Abstract: Pumpkin has long been used as traditional health materials in oriental medicine, pharmacy, medicine, and food industries in many countries. In this study, the water extract and two active components from tendril of young C. moschata Duch. were investigated on inflammation inhibitory activity. The water extract of young C. moschata Duch. showed high cell viability over 95% and it decreased the production of interlukin 6 (IL-6) and tumor necrosis factor (TNF-α) in the capacity of mouse bone marrow-derived macrop… Show more

“…They recently reported that dehydrodicaniferyl alcohol isolated from pumpkin stems inhibited adipogenic and lipogenic effects in 3T3-cells and primary mouse embryonic fibroblasts [13] and prevented ovariectomy-induced bone loss by inhibiting osteoclast differentiation [14,15]. Pumpkin tendril has also been found to inhibit inflammation, with rutin being identified as an anti-inflammatory compound in this material [16]. Recently, we reported that the water extract of pumpkin tendrils suppressed RANKL-induced osteoclast differentiation via downregulation of p38 and extracellular-signal-regulated kinase (ERK) [17].…”

“…RANKL induced the development of multinucleated osteoclasts from precursors, and TCMD treatments exerted preventive effects on the formation of the TRAP-positive osteoclast without any cytotoxicity, as well as actin ring formation and bone resorption. Our previous study reported that rutin was abundant in the extract of TCMD, which had an anti-inflammatory effect [21]. Additionally, a recent study indicated that rutin had preventive effects against RANKL-induced osteoclast differentiation by inhibiting ROS production [22].…”

“…However, pre-treatment of cells with TCMD had no inhibitory effect on IL-6 release ( Figure 1D-1F). A previous study revealed that rutin is a purified active compound in water extract of TCMD [36]. Accordingly, we further investigated whether rutin influences IL-1β secretion.…”

“…Among them, dehydrodiconiferyl alcohol (DHCA), a lignan compound isolated from C. moschata, was suggested to exhibit anti-inflammatory effects by reducing NO production [35]. The tendril of CMD (TCMD) water extract and the purified active compound rutin showed anti-inflammatory effects by inhibiting the production of pro-inflammatory cytokines such as IL-6 and TNF-α [36]. However, the role of TCMD in regulating inflammasome activity is unknown.…”
