Inflammaging: disturbed interplay between autophagy and inflammasomes

Abstract: Inflammaging refers to a low-grade pro-inflammatory phenotype which accompanies aging in mammals. The aging process is associated with a decline in autophagic capacity which impairs cellular housekeeping, leading to protein aggregation and accumulation of dysfunctional mitochondria which provoke reactive oxygen species (ROS) production and oxidative stress. Recent studies have clearly indicated that the ROS production induced by damaged mitochondria can stimulate intracellular danger-sensing multiprotein platf… Show more

“…185 Increase in oxidative stress and low-grade inflammation are the hallmarks of the aging process, which have been linked to caspase-1 activity. 186 In another study, improvement in memory was observed in rats injected intracerebroventricularly with a caspase-1 inhibitor (Ac-YVAD-CMK), 187 indicating that inhibition of caspase-1 can delay aging-associated pathologies.…”

“…Inflammation. At the organismal level, the age-related reduction in autophagy correlates with a variety of archetypal senescence-related symptoms including a general increase in smoldering inflammation that is referred to as "inflammaging" (102). Autophagy might facilitate the clearance of damaged (and potentially harmful) cells by maintaining effective efferocytosis as it reduces the propensity of leukocytes to produce pro-inflammatory cytokines (103)(104)(105).…”

“…The reduced capacity of autophagy with aging generates an inflammatory condition via activation of the NLRP3 inflammasome and such disturbed interplay between autophagy and inflammaosomes is linked to the pathogenesis of T2DM, obesity, and atherosclerosis 119 . Systemic impairment of autophagy by haploinsufficiency of the essential autophagy gene (Atg7 +/-) in ob/ob mice aggravated insulin resistance with increased lipid contents and inflammatory changes 120 .…”
