Inflammaging decreases adaptive and innate immune responses in mice and humans

Abstract: Both the innate and adaptive immune systems decline with age, causing greater susceptibility to infectious diseases and reduced responses to vaccination. Diseases are more severe in elderly than in young individuals and have a greater impact on health outcomes such as morbidity, disability and mortality. Aging is characterized by increased low-grade chronic inflammation, called “inflammaging”, measured by circulating levels of TNF-α, IL-6 and CRP, as well as by latent infections with viruses such as cytomegalo… Show more

“…Given that basal cell carcinoma and squamous cell carcinoma may clinically present as leg ulcers, it is imperative that a neoplastic ulcer be ruled out ( Figure 5 ) [ 72 ] . In rare cases, malignant transformation of chronic wounds may also occur (Marjolin's ulcer).…”

“…Alterations in both innate and adaptive immune cells in the aged have been noted 2,3 . In brief, the age-dependent effects on the innate immune response include diminished pathogen recognition, chemotaxis, and phagocytosis, and in adaptive immunity, declining numbers of naïve T lymphocytes and reduced cytotoxicity and antibody quality and quantity 2 . Vaccine efficacy is reduced in the elderly, as are increases in autoimmunity and cancer 2 .…”

“…Chronic inflammation strongly affects the pathogenesis of chronic and age-related diseases. With increasing age there is an enhanced incidence of a low level chronic inflammation in the absence of infection which is called inflammaging [26,27]. Among other cytokines, in particular interleukin 6 (IL-6) and tumour necrosis factor alpha (TNF-alpha) levels are elevated in this state.…”
