SUMMARYThe Federal District of Brazil (DF) lies within the Cerrado biome, where open shrubland (savannas) is interspersed with riverside gallery forests and permanent swamps (veredas). Trypanosoma cruzi-infected native triatomines occur in the area, but the enzootic transmission of trypanosomatids remains poorly characterized. A parasitological survey involving sylvatic triatomines (166 Rhodnius neglectus collected from Mauritia flexuosa palms) and small mammals (98 marsupials and 70 rodents, totaling 18 species) was conducted in 18 sites (mainly gallery forests and veredas) of the DF. Parasites were isolated, morphologically identified, and characterized by PCR of nuclear (mini-exon gene) and kinetoplast DNA (kDNA). Six R. neglectus, seven Didelphis albiventris and one Akodon cursor were infected by trypanosomes; wild reservoir infection is documented for the first time in the DF. kDNA PCR detected T. cruzi in five R. neglectus and mini-exon gene PCR revealed T. cruzi I in isolates from D. albiventris. Parasites infecting one bug yielded T. rangeli KP1+ kDNA amplicons. In spite of the occurrence of T. cruzi-infected D. albiventris (an important wild and peridomestic reservoir) and R. neglectus (a secondary vector displaying synanthropic behavior), a low-risk of human Chagas disease transmission could be expected in the DF, considering the low prevalence infection recorded in this work. The detection of T. rangeli KP1+ associated with R. neglectus in the DF widens the known range of this parasite in Brazil and reinforces the hypothesis of adaptation of T. rangeli populations (KP1+ and KP1-) to distinct evolutionary Rhodnius lineages.

Na região do Bairro da Ilha, município de Salto de Pirapora, Estado de São Paulo, Brasil, procurou-se observar a utilidade do método de Gómez-Núñez, no selecionamento das casas a serem submetidas ao expurgo seletivo pela aplicação de BHC. Dos edifícios considerados como negativos à inspeção rotineira, foram selecionados 21 nos quais foram instaladas as caixas-abrigo. O resultado revelou a presença de Triatoma infestans em 4 casas e 2 anexos. Além disso, concluiu-se a observação do ecótopo artificial para atração de triatomíneo, e o seu exame, após demolição completa, revelou a presença de 2 adultos e 198 ninfas daquela espécie. Conclui-se pela deficiência da inspeção manual, com ou sem a utilização de substância desalojante, na seleção dos edifícios para o rociado. O método de Gómez-Núñez poderá trazer valiosa contribuição para a melhoria do critério de seleção. Chama-se ainda a atenção do possível poder de dispersão ativa do T. infestans que poderá atingir valores ponderáveis.

Aware of the likelihood of a "false positive" diagnosis of house infestation when only fecal smears were found in Gó mez-Nú ñ ez boxes, Forattini et al (1969) applied the benzidine (Adler) test to detect heme residues and distinguish between the morphologically similar dejecta voided by cockroaches and triatomines, but quantitative results were not reported. In forensic chemistry, use of the benzidine test has been discontinued, because of its carcinogenic properties.…”

A Simple Method to Identify Triatomine (Hemiptera: Reduviidae) Feces in Sensing Devices Used in Vector Surveillance Programs

We successfully applied the phenolphthalin (Kastle-Meyer) test used in forensic chemistry to distinguish between feces from triatomines and other domestic arthropods in sensing devices used for vector surveillance. All black or dark brown, but not white or yellow, fecal smears from laboratory-reared or field-collected Triatoma infestans Klug, Triatoma guasayana Wydgozinsky & Abalos, Triatoma sordida Ståhl (recently revalidated as Triatoma garciabesi Carcavallo, Cichero, Martínez, Prosen & Ronderos) tested positive, whereas dejecta from cockroaches and spiders, crickets, beetles, predatory bugs, and domestic flies tested negative. Black or dark brown dejecta from female Aedes aegypti L. and Cimex lectularius L. bedbugs also tested positive. In sellsing devices installed in bedrooms of 11 houses in Amamá, rural northwestern Argentina, where neither cimicid bedbugs nor argasid ticks had been found over the years, only 62% of the black or dark brown fecal smears attributed to triatomines by a skilled observer tested phenolphthalin-positive. After insecticidal spraying, when bedroom areas were not colonized by triatomines, only 33-40% of the black or dark brown fecal smears in sensor boxes attributed to triatomines by another skilled observer tested phenolphthalin-positive. Eleven (79%) ofthe 14 houses with dubious or nontypical triatomine feces tested phenolphthalin-positive at least once during 1993-1995. Our study introduces a low-cost, simple and effective procedure for the identification of triatomine feces. The test, as a helpful adjunct to sensing devices used in triatomine surveillance, will aid in the accurate detection of infestations and the determination of the need for insecticide application.
