Infection pattern and transmission potential of chikungunya virus in two New World laboratory-adapted Aedes aegypti strains

Abstract: Chikungunya virus (CHIKV) is an emerging mosquito-borne virus belonging to the Togaviridae, which is transmitted to humans by Aedes aegypti and Ae. albopictus. We describe the infection pattern of CHIKV in two New World Ae. aegypti strains, HWE and ORL. Both mosquito strains were susceptible to the virus but showed different infection patterns in midguts and salivary glands. Even though acquisition of a bloodmeal showed moderate levels of apoptosis in midgut tissue, there was no obvious additional CHIKV-induce… Show more

“…However, midgut infection and dissemination of arboviruses are associated with the ingestion of a bloodmeal. CHIKV dissemination from the midgut has been observed as early as 24 h postoral acquisition of the virus, a time point at which the epithelial tissue of the blood-filled midgut was still overstretched (Dong et al, 2016). Thus, it appears that the virus was already disseminating from the midgut before transcriptional upregulation of Aemmp1 had reached its peak, which occurred at around 72 hpbm, a time point at which the bloodmeal was already digested.…”

“…CHIKV prevalence into saliva of the HWE increases from 20% at 2 dpi to 60% at 7 dpi, differing from the ORL strain (55% at 2 dpi to 65% at 7 dpi)[31]. Our study also demonstrates that the increase of the ZIKV BR saliva detection rate was more pronounced in the HWE infected mosquitoes (0% at 7 dpi to 50% at 14 dpi) in relation to the RED (0% at 7 dpi to 11.1% at 14 dpi) and ROCK (11.1% at 7 dpi to 22.2% at 14 dpi).…”

“…The presence of p400 in the mosquito tracheal system could indicate that it is involved in tracheal morphogenesis. As arboviruses can infect tracheal cells in A. aegypti (66), p400 could also mediate antiviral protection in these cells. Consistent with its role in hemocyte proliferation and differentiation in the fly model (51), p400 is expressed in hemocytes in A. aegypti females, with the protein localized in the nucleus of undifferentiated prohemocytes and in the nucleus and cytoplasm of differentiated hemocytes.…”
