Infection of the tracheal epithelium by infectious bronchitis virus is sialic acid dependent

Abstract: Avian Infectious bronchitis virus (IBV) is a coronavirus that infects chickens via the respiratory epithelium as primary target cells. The binding of coronaviruses to the cell surface is mediated by the viral surface protein S. Recently we demonstrated that alpha2,3-linked sialic acid serves as a receptor determinant for IBV on Vero cells and primary chicken embryo kidney cells. Here we analyze the importance of the sialic acid binding activity for the infection of tracheal organ cultures (TOCs) by different I… Show more

“…QX strain is rapidly replicating virus causing complete ciliostasis of TOCs in the third day post infection comparable to the fifth day post infection for other relatively slowing replicating viruses as 4/91 and Beaudette strains (Lukert and Packer, 1972;Hu et al, 1975;Andrade et al, 1982;Winter et al, 2008;Abd El Rahman et al, 2009). In addition, TOCs can easily prepared, cultivated and since recent research is eager to establish alternative culture systems to reduce and/or replace infectious studies involving live animals.…”

“…TOCs were prepared according to Winter et al (2008) and incubated at 37°C under rotation. Only TOCs with a ciliary activity of 100% were selected for comparing of the ciliostatic effect of isolated and reference viruses.…”

“…The TOC were prepared from 20-day-old specific pathogen free chicken embryos (Lohmann, Cuxhaven, Germany) and 26-day-old turkey embryos (Moorgut Kartzfehn, Bösel, Germany) as previously described (Winter et al, 2008). Briefly, the tracheae were dissected and cleaned from connective tissue and attached muscles and cut with a sterile microtome blade into rings of approximately 1 mm thickness.…”

“…Tracheal organ cultures (TOCs) have previously been used for examining viruses, including IBV, within an in vitro environment (Raj & Jones, 1996;Wang et al, 1997;Winter et al, 2008). Such studies have allowed for the comparison of strain virulence, analysis of infection mechanisms and virus neutralization.…”
