Infection of HTLV-III/LAV in HTLV-I-Carrying Cells MT-2 and MT-4 and Application in a Plaque Assay

Abstract: The human T-cell lines MT-2 and MT-4 carry the human T-cell leukemia virus type I (HTLV-I). When MT-2 and MT-4 were infected with HTLV-III, the probable etiologic agent of the acquired immune deficiency syndrome (AIDS), rapid cytopathogenic effects and cytotoxicity were observed that made it possible to titrate the biologically active virus in a plaque-forming assay. The cytopathogenic effects were preceded by the rapid induction and increase of HTLV-III antigens as revealed by immunofluorescence and immunopre… Show more

“…Initial virus stocks were produced by transfection of HeLa cells and virus was subsequently passaged in MT4 cells (Harada et al, 1985). For the selection of variants with increased resistance to ABT-77003, cell-free supernatants from acutely infected MT4 cells containing high levels of HIV-1 p24, as measured by the Dupont HIV p24 core profile ELISA, were used to infect MT4 cells in RPMI medium containing 10 % fetal calf serum, antibiotics and 0.33 gM-ABT-77003 (Abbott).…”

“…The CD4 + cell lines C8166 (Salahuddin et al, 1983), H9 (Popovic et al, 1984), MT2 and MT4 (Harada et al, 1985) were cultured in growth medium at 37 °C in 5% CO,~. Growth medium was RPMI 1640 supplemented with 10 % fetal calf serum and antibiotics.…”

“…This cell line was maintained in Dulbecco's modified Eagle's medium supplemented with 10 % fetal calf serum, plus antibiotics. MT-2 cells (Harada et al, 1985) were co-cultivated directly with patients' cryopreserved PBLs in RPMI 10 to obtain HIV stocks for sensitivity testing as described . Patient PBLs were also co-cultivated with donor (HIV seronegative) PBLs and DNA was extracted for PCR and DNA sequence analysis .…”
