Induction of Somatic Embryogenesis in Vulnerable Medicinal Plant Hedychium spicatum Buch-Ham ex Smith

Abstract: This protocol has been developed for somatic embryogenesis in Hedychium spicatum. Simultaneously, a method has also been developed for the production of synthetic seeds by using somatic embryos. Direct somatic embryos were developed on cotyledon explants of zygotic embryos on MS supplemented with high concentration of NAA (20.0 μM). Induction of secondary embryogenesis was best in 2,4-D supplemented medium fortified with activated charcoal. Germination of somatic embryos was enhanced by using GA 3 . Besides th… Show more

“…Somatic embryogenesis has already been reported in many endangered plants for their in vitro conservation (Lee et al, 2011;Kim et al, 2012;Giri et al, 2013;Barberini et al, 2016). In the present study, cytokinins were used as promoters for the induction of somatic embryos.…”

“…TDZ induces the expression of speci c genes and transcription regulatory sequences that are either directly responsible for callus formation or induce other auxins or cytokinins for callus induction (Galán-Ávila et al 2020;Ali et al 2022). The higher concentration of NAA (20 µM) showed a better response to induce direct embryogenesis from the seed radicle of the zygotic embryo (Giri and Tamta 2013).However, a commercial-scale propagation protocol has not been developed so far. Previous studies attempted to optimize PGRs concentration for micropropagation of H.spicatum through shoot tip and rhizome bud explants (Koul et al 2005;Badoni et al 2010).…”

“…Thus, the method can be used for continuous, sustainable, economical and viable production of natural compounds, regardless of environmental conditions (Karuppusamy 2009). The in vitro propagation methods are available for the species (Badoni et al 2010;Giri and Tamta, 2011;2013); however, the propagation protocol of the species at the commercial level has yet to be developed.…”

“…Shoot elongation and higher shooting percentage were also reported in MS medium supplemented with 5.0 m m kinetin and 1.0 m m IAA . Direct somatic embryos were developed by Giri and Tanta (2013) on cotyledon explants of zygotic embryos on MS supplemented with a high concentration of NAA (20.0 μ m ) . Induction of high frequency shoot multiplication (83.33%), number of shoots per explant (3.86 shoots) and average number of shoots per flask (19.33 shoots) were reported in selected cytokines combinations when seed raised shoot tips of the species were taken as explants .…”
