Induction of Photoallergy in Guinea-Pigs by Injection of Photoallergen-Protein Conjugates

Abstract: Photoconjugates were prepared by ultraviolet irradiation of guinea-pig albumin (GPA) with the photoallergens tetrachlorosalicylanilide (T4CS) and fentichlor. Injections of T4CS-GPA induced photoallergy to T4CS in 11 of 12 guinea-pigs whereas injections of fentichlor-GPA induced photoallergy in 5 of 12 guinea-pigs. Thus the fentichlor-GPA photoconjugate, which contained a molar ratio of hapten to protein 3 times higher than the corresponding T4CS conjugate, produced a… Show more

“…Recent in vivo evidence for the importance of protein-photoallergen conjugates has been generated in this laboratory [31]. From consideration of the work by Herman and Sams [30], together with the high binding affinity shown by T4CS towards serum albumin, it was argued that a photoallergic response to T4CS ought to be induced in guinea-pigs by injection of T4CS-guinea-pig albumin photoconjugate (as opposed to the conventional induction route of topical application of T4CS followed by ultraviolet irradiation).…”

“…Photoallergy resembles contact hypersensitivity; the only difference is that the drug requires activation by light, instead of, for example, enzymes, in order to provoke this unwanted immune response . The photoexcited drug or its reactive photoproduct, called hapten, irreversibly binds to proteins, resulting in the formation of an antigen; this is supposed to be the initial step in photoallergy . Subsequently, the antigen triggers immunologic reactions that eventually result in the symptoms of contact hypersensitivity, in this case, photoallergy , .…”
