Mitochondrial ion transport has been studied by many investigators since the early work of Bartley and Davies (1). Passive ion extrusion was observed on aged mitochondria or on fresh mitochondria with valinomycin (2-5). On the other hand, metabolism de pendent ion retention (6-3), ion uptake (9-17) and ion extrusion (18-20) have been found on mitochondria. It is suggested that there are some unknown factors which control ion movement in the mitochondria.Presently, in order to clarify the factor, movement of potassium and magnesium was studied in the mitochondria of the kidney cortex and liver of rats. A metabolism de pendent potassium and magnesium ion extrusion was observed in the presence of a res piratory substrate, and this extrusion was converted to retention by further addition of 1.0 mm of ATP. This effect of exogeneous ATP was disccussed.

These K + movements have not all been thoroughly characterized, but they appear to occur in association with an H + countermovement and anion movement and can take place against the chemical gradient through utilization of energy derived from oxidizable substrates or ATP.…”

The addition of agents that uncouple electron transfer from energy conservation (uncouplers) to state 4 mitochondria causes the following ion movements: K(+) is extruded from the mitochondria in association with phosphate and possibly other anions, but not H(+). Endogenous Ca(++) is extruded from the mitochondria, and H(+) moves in to counter-balance the Ca(++) movement; some phosphate movement may be associated with Ca(++) extrusion. The rate and extent of K(+) extrusion induced by uncoupler is dependent on the concentrations of external phosphate and divalent ions. Phosphate induces K(+) extrusion, while Mg(++) and Mn(++) inhibit it. TheV max of K(+) transport is 300 μmoles K(+)/g protein per min. The K m for FCCP-induced potassium extrusion is 0.25 μM at pH 7.4. The inhibitory effect of Mg(++) is noncompetitive with respect to uncoupler concentration but competitive with respect to phosphate concentration. The experimental evidence does not support the existence of high H(+) permeability in the presence of uncoupler. A correlation is observed between the rate of K(+) extrusion and the energy reserves supplied from the high energy intermediate. The action of uncoupler in inducing K(+) permeability is considered to arise through its action in depleting the energy reserves of mitochondria rather than through a specific activating effect of permeability by the uncoupler itself. The relationship of membrane potential to regulation of K(+) permeability is discussed.

SummaryBecause of the controversy among reports as to whether dietary fiber interferes with mineral absorption or not, we investigated the effect of dietary citrus pectin in a low-caloric, processed formula on serum calcium (Ca), zinc (Zn), copper (Cu), and iron (Fe) in 14 healthy obese Egyptian women undergoing weight reduction over a four-week period. The total caloric supply was 1,000 kcal/day. There was no difference between pre-and post-study values in the serum Ca. This was ascribed to the fact that water-soluble dietary fibers are fermented in the colon with the generation of short-chain fatty acids, which besides being absorbed, also enhance colonic divalent cation absorption. There was a numerical non significant decrease well within the reference values of Zn (-4.87%), Cu(-6.42%), and Fe(-19.65%). These results were discussed in light of the suggestion that the Zn, Cu, and Fe status is one aspect of an integrated acute-phase response. The response might have been triggered earlier by obesity, and later on augmented by the nutritional and psychological stress superimposed by the slimming diet. Hence, we conclude that dieters should be supplemented with minerals to replenish the body resources before embarking on low-energy diets. These diets should be of short duration, and must be interrupted by periods of normal alimentation if the need calls for their repetition.
