Induction of epithelial‐mesenchymal transition with O‐glycosylated oncofetal fibronectin

Abstract: Epithelial-mesenchymal transition (EMT) has been shown to play a key role in embryogenesis and cancer progression. We previously found that fibronectin (FN) carrying O-GalNAc at a specific site is selectively expressed in cancer and fetal cells/tissues, and termed oncofetal FN (onfFN). Here, we show that (i) a newly-established monoclonal antibody against FN lacking the O-GalNAc, termed normalFN (norFN), is useful for isolation of onfFN, (ii) onfFN, but not norFN, can induce EMT in human lung carcinoma cells, … Show more

“…This opposite expression in primary and advanced tumors indicates a dual function at different stages of cancer. These miRNAs were found to be correlated with O-glycan biosynthesis; O-glycosylation has an important functional role in EMT induction (25,26). In line with these results, we found that genes that are positively correlated with P5 miRNAs are enriched by genes that are upregulated during EMT (ref.…”

“…A switch from E-cadherin to N-cadherin expression is of strong independent importance for the progress of prostate cancer [26][27][28][29] , and up-regulation of vimentin expression is linked to prostate cancer invasion [29] . Our findings add to the growing evidence linking glycosylation and the EMT transition [30][31][32][33] , and provide important insight into understanding how O-linked glycans can influence this process. Further dissection of the interplay between sTn and oncogenic features will require deciphering the effects of sTn on additional specific O-glycoproteins.…”

“…Cancer cells that undergo EMT upregulate FN along with other ECM molecules that constitute the remodeled ECM. Interestingly, secreted FN further stimulate EMT, for example, by sensitizing cancers to TGFβ induction, and trigger a variety of signaling pathways that in turn upregulate FN expression 14, 15 (Figure 2B). This feedback loop once again reflects a close alliance of FN and cancer cells.…”
