Induction of Cell-Mediated Immunity toStaphylococcus aureusin the Mouse Mammary Gland by Local Immunization with a Live Attenuated Mutant

Abstract: The efficacy of intramammary (Ima) immunization with a live attenuated (la) Staphylococcus aureus mutant to protect the mouse mammary gland from infection has previously been established. The present study was aimed at evaluating whether Ima immunization with la-S. aureus can induce cell-mediated immune responses to the pathogen within the mammary gland. Mice were immunized by Ima route with la-S. aureus, and regional lymph node mononuclear cells were obtained thereafter. A higher expression of the interleukin… Show more

“…Sordillo and Nickerson, 1986). The murine model for the study of bovine mastitis has also been utilized to evaluate the efficacy of numerous vaccines (Mamo et al, 2000;Gomez et al, 2002), antimicrobials (Brouillette et al, 2004) and immunomodulatory compounds (Sordillo and Nickerson, 1986;Lee et al, 2003;Boulanger et al, 2007) in order to elucidate agents that might prevent or treat bovine mastitis.…”

“…Moreover, ima immunization of mice with the S. aureus aroA mutant induced high levels of both IFN-␥ and IL-4 transcripts in the mammary gland. In previous studies, we have demonstrated the feasibility of inducing Th1 and Th2 responses against S. aureus in the mouse mammary gland by local immunization with temperature-sensitive mutants of S. aureus during late pregnancy (22).…”

“…For example, cell-mediated and innate immune responses have been shown to play vital roles in conferring protection in several nonbiofilm infection models (1,18,21,23,27,28,32). Also, in other studies, neutrophils are the dominant cell type elicited during S. aureus infection and play a key role in protection against nonbiofilm infections (30,38,39).…”
