Induction of apoptotic cell death by pancreatitis‐associated ascitic fluid in Madin‐Darby canine kidney cells

Abstract: We investigated the cytotoxicity on Madin-Darby canine kidney (MDCK) cells of pancreatitis-associated ascitic fluid (PAAF) collected from rats with experimental necrotizing pancreatitis. PAAF reduced viability of MDCK cells in a time-and dose-dependent manner. We detected DNA fragmentation on the PAAF-treated MDCK cells, indicating that the cytocidal action of PAAF is via apoptosis. From the results obtained, we conclude that PAAF contains factor(s) inducing apoptosis on MDCK cells, and we assume that apoptoti… Show more

“…On the other hand, we have reported that ascitic fluid associated with experimental necrotizing pancreatitis contains the activity inducing apoptotic cell death of Madin-Darby canine kidney cells [20]. Although the factor(s) inducing apoptotic cell death in that in vitro system has not been identified, TNF-␣ secreted from activated peritoneal macrophages undoubtedly is a major candidate.…”

“…To test this hypothesis, we estimate the effect of peritoneal lavage prior to the development of pancreatitis on the cytotoxicity in the PAAF, using Madin-Darby canine kidney (MDCK) cells as target cells as reported previously [11]. In the present study, the involvement of TNF-␣ in the cytotoxicity of the PAAF is also tested and discussed.…”

“…Satake et al [10] reported that the PAAF from the canine pancreatitis model caused acute renal failure, suggesting the existence of nephrotoxic substance(s) in the PAAF. Furthermore, we reported that the PAAF from a rat necrotizing pancreatitis model had a lethal effect on cultured cell lines via apoptosis [11].…”
