The aim of this work was to study the short-term effects of salt stress on the antioxidant system and ascorbate peroxidase (APX) expression in two salt sensitive sweet potato cultivars Tainung 57 (TN57) and Tainung 66 (TN66), and one salt-tolerant cultivar Hsusu 18 (HS18). Plants were grown in plastic pots in a greenhouse for 30 d followed by NaCl treatments (0, 150, 300, and 450 mM) for 0, 24 and 48 h in a growth chamber. Young, fully expanded leaves of each treatment and period of time were clipped for enzyme activity measurements. In addition, different tissues (leaves, stems, and roots) were also harvested to analyze the tissue-specific APX gene expression using semiquantitative reverse-transcription polymerase chain reaction (RT-PCR). Three degenerated primers of APX isoforms from cytosol, peroxisomes and chloroplasts were used to amplify the APX complementary DNA of these cultivars. Our results show higher increase in APX activity at 24 and 48 h of salinity (450 mM of NaCl) in salt-stress tolerant genotype than in saltsensitive ones. The expression of APX isoforms in response to salinity was tissue specific and also dependent on stress duration.Additional key words: antioxidant enzyme, APX isoforms, Ipomoea batatas, RT-PCR.

Treatment with solutions containing high concentrations of NaCl (200 or 300 mM) induced cell death in rice (Oryza sativa L.) roots, as well as the application of exogenous hydrogen peroxide (H2O2). Moreover, the pretreatment with dimethylthiourea (DMTU), a scavenger of H2O2, partially alleviated the root cell death induced by 200 mM NaCl. These observations suggest that the cell death of rice roots under high salt stress is linked to H2O2 accumulation in vivo. NaCl stress increased the level of cyanide-resistant respiration to some extent and enhanced the transcript levels of the alternative oxidase (AOX) genes AOX1a and AOX1b in rice roots. High-salt-stressed (200 mM NaCl) rice roots pretreated with 1 mM salicylhydroxamic acid (SHAM), a specifi c inhibitor of alternative oxidase, exhibited higher levels of cell death and H2O2 production than roots subjected to either 200 mM NaCl stress or SHAM treatment alone. These results suggest that cyanide-resistant respiration could play a role in mediating root cell death under high salt stress. Furthermore, this function of cyanideresistant respiration could relate to its ability to reduce the generation of H2O2

Treatment with 300 mM NaCl increased the capacity of the alternative respiratory pathway and induced the expression of AOX1a of the leaves of rice (Oryza sativa L.) seedlings. A significant increase in the content of hydrogen peroxide (H2O2) was observed in rice leaves treated with 300 mM NaCl. However, NaCl at 150 mM did not significantly affect the capacity of the alternative respiratory pathway, the content of H2O2, and the transcript level of AOX1a. Exogenous application of H2O2 enhanced the levels of the capacity of the alternative respiratory pathway and AOX1a expression. The accumulation of H2O2 in rice leaves in response to 300 mM NaCl was inhibited by the pretreatment with dimethylthiourea (DMTU, scavenger of H2O2). This treatment also suppressed the induction of AOX1a expression and the increase in the capacity of the alternative respiratory pathway under 300 mM NaCl stress. Moreover, the salt-stressed (300 mM NaCl) seedlings pretreated with 1 mM salicylhydroxamic acid (SHAM, a special inhibitor of alternative oxidase) had higher level of H2O2 production than the seedlings either subjected to 300 mM NaCl stress or SHAM treatment alone did. These observations suggest that the expression of AOX1a in response to higher salt stress is mediated through an accumulation of H2O2 and alternative oxidase could play a role in antioxidant protection under the condition of higher salt stress.
