Inducible LGALS3BP/90K activates antiviral innate immune responses by targeting TRAF6 and TRAF3 complex

Abstract: The galectin 3 binding protein (LGALS3BP, also known as 90K) is a ubiquitous multifunctional secreted glycoprotein originally identified in cancer progression. It remains unclear how 90K functions in innate immunity during viral infections. In this study, we found that viral infections resulted in elevated levels of 90K. Further studies demonstrated that 90K expression suppressed virus replication by inducing IFN and pro-inflammatory cytokine production. Upon investigating the mechanisms behind this event, we … Show more

“…Gal-3BP is suggested to be a major upstream molecule in the activation of inflammatory factors, including IL-6 and TNF-α, by playing an important role in macrophage polarization and activation. In addition, several investigations have suggested a strong link between Gal-3BP levels and metabolic disorders, consistent with systemic pro-inflammation, by influencing inflammatory signalling pathways, including JNK-STAT and IKKβ/NF-κB [20][21][22][23]. There is solid evidence that insulin resistance is related to inflammation with both the fluctuation of cytokines and the relevant signalling pathways.…”

“…Both have been confirmed to be expressed in upper airway tissues ( Fink et al, 2013 ; Clifford et al, 2016 ). As for their respective capacities for distinguishing upper airway samples from different subjects under various pathological conditions, LAGLS3BP has a specific expression level following the activation of neutrophil-mediated immune responses, which is generally observed during viral infections ( Andres-Terre et al, 2015 ; Xu et al, 2019 ) including but not restricted to SARS-CoV-2 infection ( Didangelos, 2020 ; Park et al, 2020 ). Therefore, LAGLS3BP may also help in distinguishing samples from disease controls and those from patients with viral infection but not the detailed subgrouping of infections.…”

“…It also plays regulatory roles in the immune system; data from the literature concerning the role of 90 K in the immune response has indicated that it may have immunosuppressive as well as immunostimulatory functions depending on the biological context, and it may induce or suppress the secretion of cytokines that play roles in inflammation 1 . It is also induced during viral infections leading to IFN and pro-inflammatory response 33 . 90 K was also suggested as potential biomarker in several diseases.…”
