Individuals with inherited chromosomally integrated human herpes virus 6 (ciHHV-6) have functionally active HHV-6 specific T-cell immunity

Abstract: To evaluate the human herpes virus 6 (HHV-6) -specific immune response in individuals with chromosomally integrated HHV-6 (ciHHV-6), we measured HHV-6-antigen-specific cytokine responses (interferon-γ, interleukin-2, tumour necrosis factor-α) in T cells by flow cytometry in 12 and 16 individuals with and without ciHHV-6, respectively. All individuals with ciHHV-6 showed HHV-6-specific T cells with higher frequencies of HHV-6-specific CD8(+) cells (0.03-14.93, median 2.15% of CD8(+) cells) compared with non-ciH… Show more

“…The absence of reactivation in all inherited ciHHV-6A-positive recipients who received ciHHV-6-negative donor cells (n ϭ 6) generates speculation about whether there may be an immune response in inherited ciHHV-6-positive individuals that reduces the risk of reactivation of latent HHV-6B. Along those lines, recent work showed increased HHV-6-specific functionally active immune responses in individuals with inherited ciHHV-6 (17).…”

“…33 However, HHV-6-antigen-specific cytokine responses can be detected in individuals with iciHHV-6, suggesting the condition may not be immunologically silent. 34 Our understanding of the immunologic effects of prior herpesvirus infections continues to evolve. A large study investigating the effect of Epstein-Barr virus seropositivity on outcomes in HCT recipients with acute leukemia found an association between donor seropositivity and acute and chronic GVHD they attribute to Epstein-Barr virus-infected donor B-cell activity.…”

“…Peptide epitopes confirmed here for U14, U90, and U95 had sequence homology to the related betaherpesviruses HHV-6A (all epitopes) and HHV-7 (nine epitopes) (see Table S2 in the supplemental material), suggesting possible cross-reactivity. Twenty-one epitopes had identity with the iciHHV-6B genomic sequence (GenBank accession number KY315520.1), and albeit beyond the purview of this study, it would be interesting to test if these epitopes are recognized by HHV-6B-specific T cells in a cohort with iciHHV-6B (47)(48)(49).…”
