Individual variation in the frequency of HLA class II‐specific cytotoxic T lymphocyte precursors

Abstract: The frequencies of HLA class II-specific cytotoxic T lymphocyte precursors (CTLp) were studied in number of unrelated individuals using a limiting dilution analysis system optimized for the detection of CD4+ CTLp. Peripheral blood mononuclear cells (PBMC) were enriched for CD4+ T cells by immunomagnetic depletion of CD8+ T cells. In some allogeneic combinations high CTLp frequencies were obtained with no significant difference between PBMC and CD4-enriched PBMC populations. In other combinations CTLp frequenci… Show more

“…This suggests that both T-cell subsets can be redirected to become cytolytic against CLL cells in the presence of blinatumomab. Although cytotoxic CD4 + T cells are relatively rare in healthy subjects, 30 an expanded population of CD4 + perforin + T-cells with an EM phenotype has been described in CLL patients. 31 Our study used 3-or 7-day cultures of PBMC treated with blinatumomab.…”

“…Over the past 2 decades, the ability of CD4 ϩ T cells to display cytotoxic potential has been reported in isolated, but repeated, reports in both mouse and human (2)(3)(4)(5)(6). However, the observation of such cytotoxic activity in CD4 ϩ T cells has usually been restricted to cell lines and CD4 ϩ T cell clones generated by long term in vitro culture and has therefore been considered by some authors to be an artifact (7).…”

“…Autologous CD8 ϩ -enriched responder T lymphocytes were prepared from the nonadherent cell fraction obtained on day 0 by immunomagnetic beads (Dynal Biotech, Wirral, U.K.) and depletion of CD4 ϩ and CD16 ϩ cells (37), and then cryopreserved. On day 6, the cells were thawed and added at 2 ϫ 10 6 cells/well to the adenovirus-infected DCs (for a responder-stimulator ratio of 20:1) in 1 ml/well RPMI/AB supplemented with human rIL-7 (Genzyme, Cambridge, MA) at a final concentration of 5 ng/ml.…”
