Objective: The objective of the study was to develop and validate a simple and reproducible reverse phase high pressure liquid chromatography (RP-HPLC) method for hydrophilic drug isoniazid (INH) to apply for the analysis of the INH in nanoparticle drug formulations. Furthermore, to estimate simultaneously rifampicin (RIF) and INH in combined form. Methods:Isocratic elution with 10 minutes runtime on a C-18 Luna, 5 µ, 100Å, 150 mm column, methanol, and water as mobile phase with detection wavelength at 268 nm was used. INH nanoformulations were prepared by double emulsion solvent evaporation technique. Quantitative analysis of encapsulated drug was estimated via developed RP-HPLC method. Simultaneous estimation for the two drugs was carried out by gradient elution. All chromatographic separation and estimations were obtained on Shimadzu HPLC system. Results:INH eluted with a short retention time (RT) of 4.06 minutes. Method showed good linearity in the range of concentrations 0.01-100 µg/ml. The limit of detection (LOD) and quantification (LOQ) for INH was 0.03 and 0.12 µg/mL, respectively, and developed method has been successfully applied for the analysis of drugs in nanoparticle formulations. Simultaneous estimation of antitubercular drugs INH and RIF showed two separate peaks within specified runtime. Conclusion:Developed method showed good resolved peaks. Since the RT is short, in a shorter duration more samples could be completed and developed method will be easy for analyzing greater number of samples. Analysis of nanoformulation results have shown that this method is simple, reliable, reproducible, hence can be applied for drug delivery analysis.

Abstract:The H-point standard addition method (HPSAM) has been applied for simultaneous determination of Zn(II) and Ni(II) at trace levels, using Alizarin Red S (ARS) as chromogenic reagent. Both Ni(II) and Zn(II) form red colored complex with Alizarin red S at pH 7.0 which are soluble in triton x-100. Zn(II) and Ni(II) have been determined simultaneously in the concentration range of 0.320-4.55 µg/mL and 0.293-4.676 µg/mL respectively with satisfactory accuracy and precision. The results of applying H-point standard addition method show that Zn(II) and Ni(II) ions can be determined simultaneously with concentration ratio of 15:1 and 1:16 of Zn(II) to Ni(II). This method is based on the difference in the absorbance of Zn(II) and Ni(II) -ARS complexes at pH 7.0 using different wavelength pairs.

IntroduçãoA hidrazina é amplamente usada na indús-tria e na farmacologia como inibidor, catalisador, corante e combustível. Na indústria é usada como agente desincrustante de resíduos de caldeiras. Devido à elevada temperatura observada em águas de caldeira, determinados compostos tais como: íons cálcio, magnésio, silicatos, sulfatos e carbonatos, que são mantidos solubilizados em água de alimentação/reposição, tendem-se a depositar sobre a superfície da caldeira face á redução de sua solubilidade. Outros metais tais como ferro, zinco, cobre e chumbo podem também constribuir para incrustações nas paredes das caldeiras, provocando grandes prejuízos econômicos. Várias ações corretivas e preventivas destas incrustações são utilizadas pelas indústrias, incluindo adição de hidrazina, polímeros, controle da alcalinidade ou agentes sequestrantes de oxigênio dissolvido.A ação da hidrazina no interior das caldeiras é de agente redutor, que atua na desaeração química das águas em alta temperatura, reduz a corrosão galvânica e elimina a contaminação da água por cobre metálico. Ela também é explorada como maté-ria-prima para muitos derivados, tais como agentes espumantes para plásticos, antioxidantes, pesticidas, fertilizantes e fármacos [1,2]. No entanto, seu uso tem sido desencorajado, pois a hidrazina apresenta alta toxicidade, podendo apresentar características carcinogênicas e mutagênicas. Deste modo métodos analíticos sensíveis e rápidos para sua detecção e determinação em águas residuais provenientes de vários tipos de indústria e outros efluentes, tem sido requeridos para atender aos órgãos fiscalizadores.A literatura reporta vários métodos analíti-cos para determinação de hidrazina. Dentre eles podemos destacar os métodos cromatográficos [3,4] Resumo: Eletrodos de carbono vítreo foram modificados com filmes de poli ácido glutâmico (PAG) pela eletropolimerização do ácido glutâmico em tampão fosfato pH 7,0, utilizando-se varreduras sucessivas de potencial entre -0,8 a +2,0 V. Os eletrodos modificados foram aplicados na determinação de hidrazina, a qual é pré-concentrada na superfície através da interação entre grupos carboxílicos livres no filme e grupo amino da hidrazina. Embora a hidrazina não seja oxidada em intervalo útil de potencial sobre eletrodo convencional, no eletrodo modificado caracteriza-se pelo pico de oxidação em potencial de +0,57 V usado para fins analíti-cos. Gráfico de calibração linear foi obtido no intervalo de concentração entre 4 x 10 -5 a 1 x 10 -3 mol L -1 , usando voltametria de pulso diferencial. O limite de detecção foi de 1,2 x 10 -6 mol L -1 . O método foi aplicado para a determinação de hidrazina em amostras coletadas de água de caldeira com recuperação de 94%.Palavras-chave: eletrodo modificado; filme de poli ácido glutâmico; hidrazina.
