The goal of this study was to use bacteriocin from a probiotic strain of Lactobacillus pentosus S6 (KU92122) to synthesize innovative strong silver nanoparticles (AgNPs). UV-Vis spectroscopy was used to analyze the synthesized nanoparticles, and the maximum absorbance was found to be approximately 450 nm. AgNPs produced were spherical in shape, with an average size of 50 nm, as validated by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Fourier transform infrared spectroscopy (FTIR) was used to confirm the presence of distinct functional groups on the surface of nanoparticles. The positive antibacterial activity of produced silver nanoparticles was tested against multidrug resistant bacteria such as Staphylococcus aureus and Bacillus cereus, as well as challenging plant pathogens such as Aspergillus niger, Pythium aphanidermatum, Fusarium oxysporum, and Phytopthora parasitica with an apparent aim to use these AgNPs with bacteriocins as effective antimicrobial agents that can be employed at broader scale in clinical and biocontrol applications especially in medicine, agriculture and the food processing sectors.

In Silico and In Vitro Evaluation of the Antimicrobial Potential of Bacillus cereus Isolated from Apis dorsata Gut against Neisseria gonorrhoeae

Antimicrobial resistance is a major public health and development concern on a global scale. The increasing resistance of the pathogenic bacteria Neisseria gonorrhoeae to antibiotics necessitates efforts to identify potential alternative antibiotics from nature, including insects, which are already recognized as a source of natural antibiotics by the scientific community. This study aimed to determine the potential of components of gut-associated bacteria isolated from Apis dorsata, an Asian giant honeybee, as an antibacterial against N. gonorrhoeae by in vitro and in silico methods as an initial process in the stage of new drug discovery. The identified gut-associated bacteria of A. dorsata included Acinetobacter indicus and Bacillus cereus with 100% identity to referenced bacteria from GenBank. Cell-free culture supernatants (CFCS) of B. cereus had a very strong antibacterial activity against N. gonorrhoeae in an in vitro antibacterial testing. Meanwhile, molecular docking revealed that antimicrobial lipopeptides from B. cereus (surfactin, fengycin, and iturin A) had a comparable value of binding-free energy (BFE) with the target protein receptor for N. gonorrhoeae, namely penicillin-binding protein (PBP) 1 and PBP2 when compared with the ceftriaxone, cefixime, and doxycycline. The molecular dynamics simulation (MDS) study revealed that the surfactin remains stable at the active site of PBP2 despite the alteration of the H-bond and hydrophobic interactions. According to this finding, surfactin has the greatest antibacterial potential against PBP2 of N. gonorrhoeae.

Lactic acid bacteria have been advocated as probiotics to replace antibiotic growth promoters, improve growth performance, and reduce cost production in swine farming. The aim of this study is to identify and evaluate the probiotic traits of strain LCN13 isolated from traditionally fermented feed for swine. Thirty-five bacterial isolates with different morphological characteristics were isolated, among which 9 isolates showed the ability to produce lactic acid and antibacterial activity against pathogenic bacteria. Among them, isolate LCN13 exhibited a strong capacity to produce lactic acid (18.5 ± 0.31 g/L), inhibited gastrointestinal pathogens such as Salmonella typhimurium ATCC 14028 (18.3 ± 1.52 mm), Escherichia coli ATCC 11105 (24.7 ± 2.14 mm), and Staphylococcus epidermidis ATCC 35984 (31.6 ± 2.93 mm), and produced 182 ng/mL folate and 233 ng/mL riboflavin as measured by LC-MS/MS after 24 hours of incubation. Based on morphological, biochemical, and 16S rRNA gene analysis, the isolate LCN13 was identified as Lactobacillus plantarum. Phenotypic analysis revealed that L. plantarum LCN13 showed remarkable resistance to 1.2% ox-bile salt, 2.0 mM H2O2, and pH 3.0. In addition, the ability to produce high levels of folate (253.6 ± 10.7 ng/mL) and riboflavin (312.0 ± 12.2 ng/mL) after 48 hours was exploited for the first time in the L. plantarum. Taken together, L. plantarum LCN13 might serve as a potential probiotic candidate for animal farming.
