Increased trunk fat is associated with altered gene expression in breast tissue of normal weight women

Abstract: Increased trunk fat is associated with an elevated risk of breast cancer in normal-weight postmenopausal women. The main objective of this study was to determine whether levels of trunk fat are associated with changes in breast gene expression in normal-weight women. Non-tumorous breast tissue was collected from 32 normal BMI women who underwent mastectomy for breast cancer risk reduction or treatment. Body composition was measured by dual-energy x-ray absorptiometry. High levels of trunk fat were associated w… Show more

“…Adipokines and chronic inflammatory factors have also been implicated in obesity. Trunk fat and android fat were inversely correlated with adiponectin ( 40 , 41 ), whereas correlated with increased leptin ( 42 , 43 ). The leg fat was positively correlated with adiponectin ( 44 ).…”

“…VEGF and leptin serum levels are also dependent on IL6 gene polymorphism in children with obesity: higher values of VEGF level were found in case of the IL6 572 gene CC and GG genotypes, whereas leptin levels depended on IL6 174 and IL6 572 gene polymorphisms 105 . Elevated levels of trunk adiposity are associated with high leptin gene expression and elevated VEGF C level in WAT from breast, even in subjects without obesity 106 . In subjects with obesity, the number of mitochondrial deoxyribonucleic acid mtDNA in mononuclear blood cells has been found to correlate with leptin blood level, which also correlates with the concentration of the circulating adhesion molecule VCAM‐1.…”

“…[112][113][114][115] Studies of patients with obesity have confirmed the genetic or epigenetic aspect of leptin-VEGF crosstalk, pointing to the role of obesity-dependent leptin gene regulation. [104][105][106][107] These studies have also revealed that leptin-VEGF crosstalk is a complicated network, involving leptin canonical and noncanonical signaling pathways, HIF-1A, and NfkB as the core, 97,124,136 with a range of substances (including adiponectin, visfatin, ghrelin, IL-6, insulin, cholesterol, LDL, triglycerides, ApoB, trimethylamine N-oxide, and NO) acting as accompanying, modifying, and disrupting factors. [99][100][101] In female patients with excess body mass and related disorders, leptin-VEGF crosstalk may be involved in PCOS, adverse pregnancy outcome, 129 and preeclampsia.…”
