Increased Serum Concentration of Angiogenic Factors in Malignant Melanoma Patients Correlates With Tumor Progression and Survival

Abstract: Our data suggest that the angiogenic serum factors VEGF, bFGF, and IL-8 are useful predictive markers for overall and progression-free survival in melanoma patients.

“…In contrast, no correlation between survival and serum or plasma VEGF levels was observed. Different threshold values of serum VEGF levels have been tested, particularly 363 and 462 pg ml À1 as those values were recently described as being correlated to survival in melanoma and lymphoma (Salven et al, 2000;Ugurel et al, 2001). In this series, the median survival was 9 months for patients with VEGF levels lower than 462 pg ml À1 and 13 months for other patients (P ¼ 0.9, Figure 3).…”

“…For this analysis, we tested three different threshold values for each of the two variables: the lowest quartile, the median and the highest quartile. In addition, we tested the detection limit for IL-6 and two threshold values previously reported as relevant for VEGF (Salven et al, 2000;Ugurel et al, 2001). The Cox proportional hazard regression model was used to identify independent prognostic factors for survival.…”

“…Elevated levels of CXCL8 have been detected in variety of tumors, such as ovarian carcinoma [157], NSCLC [158], metastatic melanoma [159], and colon carcinoma [160]. Studies from our laboratory and others suggest that the expression of CXCL8 correlates positively with disease progression [161][162][163][164].…”

“…In patients with cancer, high serum concentrations of VEGF are associated with several unfavourable clinical parameters such as short tumour volume doubling time, progressive disease, extensive disease and poor survival (Dirix et al, 1996(Dirix et al, , 1997Salven et al, 1997a,b Kitamura et al, 1998Tempfer et al, 1998;Ugurel et al, 2001). Similarly, high concentrations of b-FGF have been detected in either urine or serum of cancer patients and associated with tumour bulk in head and neck or with a short tumour volume doubling time in colorectal cancer (O'Brien et al, 1995;Leunig et al, 1998;Kumar et al, 1998).…”
